    <p> Diphenylarsinic acid (DPAA), a neurotoxic organic arsenical, is present in groundwater and soil in some regions of Japan owing to illegal dumping. The present study evaluated the potential carcinogenicity of DPAA, including investigating whether bile duct hyperplasia in the liver that was observed in a chronic study on 52 week mouse, develops into a tumor when administered to mice in their drinking water for 78 weeks. DPAA was administered to 4 groups of male and female C57BL/6J mice at concentrations of 0, 6.25, 12.5, and 25 ppm in drinking water for 78 weeks. A significant decrease in the survival rate was found for females in the 25 ppm DPAA group. Body weights of males in the 25 ppm and females in the 12.5 and 25 ppm DPAA groups were significantly lower than those of the controls. Histopathological evaluation of neoplasms in all tissues showed no significant increase in tumor incidence in any organ or tissue of 6.25, 12.5, or 25 ppm DPAA-treated male or female mice. In conclusion, the present study demonstrated that DPAA is not carcinogenic to male or female C57BL/6J mice. Taken together with the fact that the toxic effect of DPAA is predominantly restricted to the central nervous system in humans, and the finding that DPAA was not carcinogenic in a previous 104-week rat carcinogenicity study, our results suggest that DPAA is unlikely to be carcinogenic in humans.</p>

    <p> Cigarette smoking is known to increase the risk of cancer and chronic obstructive pulmonary disease (COPD). In this study, we evaluated the effects of short-term nose-only inhalation exposure to cigarette smoke in mice. Male 10-week-old C57BL mice were exposed to clean air (control) or mainstream cigarette smoke for 1 h/day, 5 days/week, for 2 or 4 weeks. Exposure to cigarette smoke increased the number of inflammatory cells, especially neutrophils, in the bronchoalveolar lavage fluid, increased inflammatory cell infiltration foci, and caused an increase in the thickness of the peripheral bronchial epithelium. Microarray gene expression analysis indicated that smoke exposure induced inflammatory responses, including leukocyte migration and activation of phagocytes and myeloid cells, as early as two weeks after the initiation of exposure. Importantly, chemokine (C-C motif) ligand 17, resistin-like alpha, and lipocalin 2 were upregulated and may serve as useful markers of the toxic effects of exposure to cigarette smoke before pulmonary histological changes become evident.</p>

    <p> Phosphorylation of histone H2AX at serine 139 (γ-H2AX) is known to be induced by direct DNA damage or cellular metabolic imbalances and malfunctions. Previous studies have reported that γ-H2AX is a useful biomarker for early detection of genotoxic bladder carcinogens in rats. The purpose of the present study was to determine the role of γ-H2AX as a biomarker for detection of non-genotoxic bladder carcinogens in rats. Six-week-old male F344 rats were treated with 15 different chemicals for 4 weeks. Immunohistochemical analyses revealed that all three genotoxic bladder carcinogens and six out of seven non-genotoxic bladder carcinogens significantly increased γ-H2AX formation in the bladder urothelium of rats. In addition, four out of five rat bladder noncarcinogens did not increase γ-H2AX formation in the bladder urothelium regardless of genotoxicity. These results suggest that γ-H2AX is a useful biomarker for detection of both genotoxic and non-genotoxic bladder carcinogens in rats.</p>

    Non-alcoholic steatohepatitis (NASH) is a recognized risk factor for liver fibrosis and malignancies, and is associated with features of metabolic syndrome, such as obesity and insulin resistance (IR). We previously demonstrated that the disturbance of connexin 32 (Cx32), a gap junctional protein of hepatocytes, exacerbated NASH in Cx32 dominant-negative transgenic (Cx32ΔTg) rats fed methionine choline-deficient diet (MCDD). MCDD is well-established means of inducing NASH in rodents; however, the Cx32ΔTg-MCDD NASH model does not reproduce obesity and IR. In this study, we aimed to establish an improved NASH model. Eight-week-old male Cx32ΔTg and wild-type (Wt) rats received a high-fat diet (HFD) with dimethylnitrosamine (DMN) for 12 weeks. The HFD with DMN led to gains in body, liver, and visceral fat weights in both genotypes. IR was significantly greater in Cx32ΔTg than in Wt rats. Elevation of serum hepatic enzymes (AST, ALT), inflammatory cytokine expressions (Tnfα, Il-6, Tgf-β1, Il-1β, Timp2, and Col1a1), steatohepatitis, and fibrosis were significantly greater in Cx32ΔTg as compared with Wt rats. Regarding carcinogenesis, the number and area of glutathione S-transferase placental form (GST-P)-positive preneoplastic hepatic foci were significantly increased in Cx32ΔTg versus Wt rats. Moreover, activation of NF-κB and JNK contributed to the progression of NASH in Cx32ΔTg rats. These results suggest that Cx32 dysfunction promoted the progression of NASH, metabolic syndrome, and carcinogenesis. Therefore, the novel Cx32ΔTg-HFD-DMN NASH model may be a rapid and useful tool for evaluating the progression of NASH.

Multi-walled carbon nanotube-7 (MWCNT-7) fibers are biopersistent and have a structure similar to asbestos. MWCNT-7 has been shown to induce malignant mesothelioma when administered by intrascrotal or intraperitoneal injection in rats and mice, and an inhalation study demonstrated that rats exposed to respirable MWCNT-7 developed lung tumors. MWCNT-N, which is similar to MWCNT-7, was shown to induce both lung tumors and malignant mesothelioma in rats when administered by trans-tracheal intrapulmonary spraying (TIPS). The present study was performed to investigate the carcinogenicity of MWCNT-7 when administered by the TIPS method. Ten-week-old male F344/Crj rats were divided into 3 groups and administered 0.5 mL vehicle, 0.250 μg/mL MWCNT-7 or 0.250 μg/mL crocidolite once a week for 12 weeks (total doses of 1.5 mg/rat) and then observed for up to 104 weeks. Rats in the MWCNT-7 group began to die from pathologies associated with the development of malignant mesothelioma 35 weeks after the final TIPS administration. Overall, the incidence of malignant mesothelioma in the MWCNT-7 group was significantly higher than in the vehicle or crocidolite groups.

    85歳男性。難治性心不全に十二指腸乳頭部癌を合併し2回目の心臓カテーテル検査で心筋生検を行いAL型心アミロイドーシスと診断、心不全3年および十二指腸癌1年という緩徐な経過で心不全死された。難治性心不全のため抗がん剤の使用歴はない。病理解剖の結果、全身性AA型アミロイドーシスと診断することができた。(著者抄録)

    われわれは前立腺癌の発生や進展にレニン-アンジオテンシン系が関係しており、アンジオテンシン2のAT1受容体ブロッカーがアンドロゲン受容体(AR)発現抑制して抗腫瘍効果があることを示した。この研究では、アンジオテンシン2のもう一つの受容体であるAT2受容体が前立腺癌の発生に関わっているか調べた。その結果、AT2受容体アゴニストのC21がAR発現抑制によって前立腺癌の発生を抑制して、化学予防効果のあることが示された。(著者抄録)

    69歳女性。メキシレチン塩酸塩内服開始約1ヵ月後より両上肢に皮疹が出現し近医を受診、接触性皮膚炎として抗アレルギー薬を処方されるも改善せず、皮疹が拡大したため、著者らの皮膚科へ紹介となった。初診時、体幹・四肢に多形紅斑を認め、3日後には皮疹が増悪し、全身倦怠感を伴ったため入院となった。入院時、両側腹部と大腿内側には暗赤色の紅斑上に粟粒大で毛孔不一致の小膿疱が散在し、膿疱は数日で消退したものの、紅斑は遷延した。経過中に発熱、リンパ節腫脹、好酸球の上昇、軽度の肝機能障害、ヒトヘルペスウイルス6型の再活性を認めたことから、本症例はメキシレチン塩酸塩による薬剤性過敏症症候群と診断された。治療としてメキシレチン塩酸塩の中止、プレドニゾロンの内服を行なったところ、紅斑は徐々に軽快し、経過良好であった。尚、本症例は発疹学的には急性汎発性発疹性膿疱症に近似しており、鑑別を要した。

    We herein report a 75-year-old female patient with intravascular lymphomatosis (IVL) who presented with fever of unknown origin. Examination, including contrast-enhanced CT and 67Ga scintigraphy, failed to show any lesions. Her blood levels of lactate dehydrogenase and soluble interleukin-2 receptors were high, suggesting a lymphomatous tumor. A bone marrow puncture was negative, and a random skin biopsy revealed a monoclonal proliferation of naked, large lymphocytes in the vascular space of the subcutaneous tissue, confirming the diagnosis of IVL. MRI, performed 7 weeks after admission, showed a brain mass mimicking primary central nervous system lymphoma. The mass was considered to be a collection of malignant lymphocyte cells invading from the vessels. Without the random skin biopsy, this case may have been misdiagnosed as primary central nervous system lymphoma. © 2014 S. Karger AG, Basel.

    症例は61歳男性で、7年前に右腎門部の腫瘍が発見され、腎摘出術で腎細胞癌と診断されていた。左側頭部腫脹と左眼球突出を自覚し、CTで蝶形骨や眼窩外側に骨融解を伴い、側頭部軟部組織へ進展した腫瘍が左中頭蓋窩前方に低吸収域として描出された。腫瘍はMRIのT1WIで辺縁高信号、内部等信号、T2WIで高信号を示し、gadoliniumで辺縁がリング状に増強された。腫瘍摘出術を施行し、病理組織所見で腫瘍中心部は広範な凝固壊死を示し、辺縁に骨や筋肉へ浸潤した腫瘍細胞が認められた。比較的均一な核を有する円形〜類円形の腫瘍細胞が、毛細血管と単層で紡錘形のsustentacular cellとで分葉されて胞巣状に配列し、変性を認めたが核異型、核分裂像、脈管浸潤はなかった。既往の腎門部腫瘍と同様の所見であり、paragangliomaの頭蓋骨転移と診断された。術後症状は改善し化学療法を追加したが、2年2ヵ月後より肩甲骨などに転移を生じ、状態は徐々に悪化して4年1ヵ月後に死亡した。

    Transgenic (TG) rats bearing a probasin promoter/simian virus 40 T antigen (SV40 Tag) construct were treated with antiandrogens to examine their ability to suppress prostate carcinogenesis. Finasteride and flutamide were administered to 10-week-old TG rats five times a week for 2, 5 and 7 weeks. Antiandrogen-treated prostates exhibited atrophic glandular structures with almost no expression of SV40 Tag and only weak signals for androgen receptors. Furthermore, quantitative data for ventral prostate adenocarcinomas showed significant decrease with antiandrogen treatment. Both finasteride and flutamide had the ability to suppress SV40 Tag-driven carcinogenesis through their different antiandrogenic mechanisms, suggesting that this TG model is suitable for exploring the potential of agents to inhibit prostate cancer development.

    ラット肝細胞初代培養法で発現変化を検討した。24時間後、48時間後と対照であるDMSO(final concentration 0.1%)曝露では形態学的に著変ないものの、Aflatoxin B1を曝露した細胞では変性を伴った。検討可能な遺伝子は20128存在し、その遺伝子を対象にMicroarrayの解析方法より、発癌物質と非発癌物質間で発現の異なる遺伝子10066が選出され、4種の発癌物質で遺伝子変動が平均1.5倍以上の71遺伝子が選出された。この71遺伝子を対象に4種の非発癌物質の遺伝子発現の平均と発現差が1.5倍以上あるものは28遺伝子であった。

    48歳男.Cellular angiofibromaと考えられた右大腿部軟部腫瘍症例であった.約4年前から右大腿部の無痛腫瘤を自覚し,徐々に増大した.初診時は6×6cm大となっており,MRIでは縫工筋内に,MRI T1強調像で筋肉と等信号,T2強調像で不均一な高信号を示す腫瘍を認め,低信号の被膜様構造も認めた.生検を行い,病理学的には小血管の豊富な,myxomatousな間質を持つ腫瘍であった.臨床的には悪性と考え広範切除を行い,術後8ヵ月現在,再発は認められない.腫瘍は黄白色の充実性で,隣接する筋肉への浸潤を示す部分も認められ,組織学的には浮腫状の間質の中に多数の壁の薄い血管腔がみられた.免疫組織化学的にはdesmin陽性,SM actinおよびHHF35は部分的陽性,S-100,CD31,CD34,EMAは陰性であった

    転移性骨腫瘍の疑いの34例にCTガイド下針生検を行った.正診率は100%であり,合併症は認めなかった.腫瘍細胞が同定されたのは22例で,そのうち14例は採取された組織結果により,原発巣やや癌種の診断が可能であった.4例は原発巣や癌種を診断できず,免疫組織化学染色により2例が肺の腺癌と診断された.CTガイド下針生検は適応症例を慎重に選ぶことが必要であり,原発不明癌の原発巣探索には確実な組織採取と免疫組織化学染色による検討が必要であると思われた

    48歳男.右大腿部腫瘤を主訴とした.右大腿部の無痛性腫瘤が約4年の経過で増大傾向を示し,MRIのT1強調画像で低信号,T2強調画像で不均一な高信号を呈した.広範切除施行後は無再発で経過している.病理組織学的に浮腫状の間質の中にみられる多数の壁の薄い血管腔が特徴的であり,紡錘形細胞の密度は不均一で厚い束状の膠原線維も認めた.免疫組織化学的検索ではdesmin陽性,SM,actinとHHF35に一部陽性,S-100,CD31,CD34,EMA陰性でmyofibroblasticな分化を示すものであり,大腿部に発生したangiofibromaと診断した.WHOの分類ではcellular angiofibromaに属するものと考えられ,希少な症例として報告した

    職業に起因する発がん要因に曝露され生じるがんを職業がんといい、産業の発達とともに大きな問題となっている。職業性膀胱がんは主に染料産業に発生し、芳香族アミンが主な原因物質として報告されている。日本でも2019年にオルトトルイジンにさらされる業務による膀胱がんが、厚生労働省の「職業病リスト」に記載され、労災補償の対象となり話題となった。われわれは同工場で生産されていた新規芳香族アミンの発がん性およびそのメカニズムを、動物実験を介して解明した。ここでは、職業がんの歴史から発がん物質の検出法、動物実験モデルの有効性について解説したい。(著者抄録)

    前立腺癌発生には種々の原因が知られているが、慢性炎症もその1つと考えられている。前立腺検体における慢性炎症からみた疫学的研究では前立腺炎と癌発生との関連について一定の見解は得られていないが、実験的レベルではその相関性が明らかにされている。前立腺癌は急増する癌であり、前立腺癌予防法の確立は重要課題である。前立腺における炎症関連発癌研究の進展によりそのメカニズムが詳細に解明され、炎症を標的とした前立腺癌予防法が確立されることが期待される。(著者抄録)

    ギャップ結合は細胞間結合装置の一種で、隣接細胞間の小分子交換を介して組織や生体のホメオスタシスを担っている。当研究室では、肝細胞のギャップ結合タンパクであるCx32(connexin 32)がドミナントネガティブに抑制され、細胞間連絡能が著減したトランスジェニックラットを樹立し、肝障害や肝発癌に対するギャップ結合性細胞間コミュニケーションの役割を解析してきた。本稿では、アルコール性肝発癌および非アルコール性脂肪肝炎に関連した発癌感受性に対する細胞間コミュニケーションの影響と、それぞれの発癌に関与する遺伝子を同定した最近の知見を報告する。(著者抄録)

    ヒストンアセチル化を促進するHistone deacetylase(HDAC)阻害剤は前立腺癌を含む様々な癌腫に対する抗癌作用を有し、増殖抑制メカニズムの特異性から前立腺癌に対する新規抗癌剤として期待されている。一方、前立腺癌は発症までの期間が長いことから予防的介入が可能な癌として注目されており、HDAC阻害剤は前立腺癌の予防物質としても期待される。本章では臨床応用を考え開発が進んでいる、副作用の少ない選択的HDAC阻害剤の癌治療効果、予防効果を報告する。(著者抄録)

    前立腺癌は環境に応じた多様な増殖機構を有することから、前立腺癌の治療には多様な増殖機構を幅広く抑制する治療法の開発が必要である。ヒストンアセチル化は遺伝子のエピジェネティックな発現制御機構の1つであり、ヒストンアセチル化を促進するHistone deacetylase(HDAC)阻害剤は癌細胞に対する増殖抑制作用を有し、前立腺癌に対する新規抗癌剤の候補になり得る可能性がある。本章ではヒストンアセチル化とHDAC阻害剤の概要、およびHDAC阻害剤の前立腺癌に対する増殖抑制メカニズムを中心に報告する。(著者抄録)
