    Enterotoxigenic Escherichia coli (ETEC), one of the diarrheagenic E. coli, is the most common cause of diarrhea in developing country, and in travelers to those areas. In this study, Caenorhabditis elegans was used as an alternative model host to evaluate ETEC infections. The ETEC strain ETEC1, which was isolated from a patient with diarrhea, possessed enterotoxins STh, LT1, and EAST1 and colonization factors CS2 and CS3. Live ETEC1 shortened the lifespan and body size of C. elegans in association with increased expression of enterotoxin genes and intestinal colonization. In contrast, heat-killed ETEC1 did not affect the lifespan of C. elegans. C. elegans infected with ETEC1 showed upregulated expression of genes related to insulin-like peptides and host defense responses. These results suggest that ETEC1 exhibits pathogenicity through intestinal colonization and enterotoxin production in C. elegans. This system is useful as an ETEC infection model.

    Lactococcus lactis subsp. lactis exhibits probiotic properties in humans. Considering that Caenorhabditis elegans can be used to study the effects of microorganisms on animal behavior, owing to its simple nervous system, we assessed the impacts of two strains of Lactococcus lactis subsp. Lactis-a non-nisin-producing strain, NBRC 100933 (LL100933), and a nisin-producing strain, NBRC 12007 (LL12007)-on the lifespan, locomotion, reproductive capacity of, and lipid accumulation in, C. elegans. The lifespan of adult C. elegans fed a mixture (1:1) of Escherichia coli OP50 and LL100933 or LL12007 did not show a significant increase compared to that of the group fed a standard diet of E. coli OP50. However, the nematodes fed Lactococcus strains showed notable enhancement in their locomotion at all of the tested ages. Further, the beneficial effects of LL100933 and LL12007 were observed in the daf-16 mutants, but not in the skn-1 and pmk-1 mutants. The lipid accumulation in the worms of the Lactococcus-fed group was lower than that in the control group at all experimental ages. Overall, LL100933 and LL12007 enhance the locomotor behavior of C. elegans, likely by modulating the PMK-1/p38 MAPK and SKN-1/Nrf2 transcription factors.

    ABSTRACT

    The complete genome sequence of mycobacterial strain YM-3, isolated from cultured yellowtail in 1986, was determined. The strain was Mycobacterium pseudoshottsii , a closely related subspecies of Mycobacterium marinum , so the strain was isolated earlier than the first report of the subspecies in 2005.

    AIM: Bacillus subtilis var. natto is used in the production of natto, a typical Japanese fermented soybean food. Although the probiotic attributes and health-related effects of B. subtilis var. natto have been reported, the effect on longevity remains unknown. In the present study, the effects of B. subtilis var. natto strains on lifespan extension and the molecular mechanisms governing the prolongevity were examined using Caenorhabditis elegans as a model animal. METHODS AND RESULTS: Synchronized 3-day-old (young adult) worms were fed Escherichia coli OP50 (control) or a subcloned isolate of B. subtilis var. natto Miyagino strain (MI-OMU01) and subjected to lifespan, survival against pathogens and abiotic stress resistance assays. Notably, the lifespan of worms fed MI-OMU01 was significantly longer than that of the animals fed OP50. Moreover, MI-OMU01 increased the resistance of C. elegans to several stressors, including UV irradiation, H2O2, and Cu2+. CONCLUSIONS: Genetic and gene expression analyses using mutant animals suggested that MI-OMU01 extended the lifespan of worms in TIR-1/SARM, p38 MAPK, and insulin/IGF-1 signaling pathway-dependent manners.

    Abstract

    Background

    Since 1996, many studies have reported that periodontal disease during pregnancy may be a risk factor for preterm birth and low birth weight; however, in Africa, periodontal disease is considered a non-high-priority disease. In addition, there are few dental facilities in rural Rwanda; thus, the oral condition of pregnant women has not been investigated. The objective of this study was to assess the tooth brushing habits of pregnant women in rural Rwanda and evaluate whether periodontal bacteria in the oral cavity of pregnant women are related to birth outcomes or oral cleaning habits.

    Methods

    A questionnaire survey and saliva collection were conducted for pregnant women in the catchment area population of Mibilizi Hospital located in the western part of Rwanda. Real-time PCR was performed to quantitatively detect total bacteria and 4 species of periodontal bacteria. The relationship of the copy number of each bacterium and birth outcomes or oral cleaning habits was statistically analyzed.

    Results

    Among the participants, high copy numbers of total bacteria, Tannerella forsythia, and Treponema denticola were correlated with lower birth weight (p = 0.0032, 0.0212, 0.0288, respectively). The sex ratio at birth was higher in women who had high copy numbers of Porphyromonas gingivalis and T. denticola during pregnancy (p = 0.0268, 0.0043). Furthermore, regarding the correlation between oral cleaning habits and the amount of bacteria, the more frequently teeth were brushed, the lower the level of P. gingivalis (p = 0.0061); the more frequently the brush was replaced, the lower the levels of P. gingivalis and T. forsythia (p = 0.0153, 0.0029).

    Conclusions

    This study suggested that improving tooth brushing habits may reduce the risk of periodontal disease among pregnant women in rural Rwanda. It also indicated that the amount of bacteria is associated with various birth outcomes according to the bacterial species. Both access to dental clinics and the oral cleaning habits of pregnant women should be important considerations in efforts to alleviate reproductive-related outcomes in rural Africa.

    Abstract

    Mycobacterium avium subsp. hominissuis (MAH) is one of the most prevalent mycobacteria causing non-tuberculous mycobacterial (NTM) disease in human and animals. Of note, MAH is a major cause of mycobacterial granulomatous mesenteric lymphadenitis outbreaks in pig populations. To determine the precise source of infection of MAH in a pig farm and to clarify the epidemiological relationship among pig and human MAH lineages, we collected 50 MAH isolates from pigs reared in Japan and determined draft genome sequences of 30 isolates. A variable number of tandem repeat (VNTR) analysis revealed that most pig MAH isolates in Japan were closely related to North American, European and Russian human isolates but not to those from East Asian human. Historical recombination analysis revealed that most pig isolates could be classified into SC2/4 and SC3. To our surprise, four isolates belonged to a new lineage (SC5) in the global MAH population. Members of SC5 had few footprints of inter-lineage recombination in the genome, and carried 80 unique genes, most of which were located on lineage specific-genomic islands. As its unique genetic features, we were able to trace the putative transmission route via their host pigs. Together, these results reveal the diversity in the pig MAH population and clarify the possibility of species-specificity in MAH in addition to local adaptation. Moreover, our study also clarified the difference of source of infection and the vector of transmission between humans and pigs and provides a clear countermeasure to prevent MAH spreading in pigs.

    Importance

    Mycobacterium avium subsp. hominissuis (MAH) is a dominant cause of non- tuberculous mycobacterial (NTM) disease in human and animals. In this study, in order to clarify the epidemiological relatedness between MAH isolates from human and pigs, and the MAH transmission route among pigs, we conducted molecular and genomic- based epidemiological research. The pig isolates were genetically distant from those of humans in Japan, however, they were closely related to human isolates from North America, Europe and Russia, suggesting a proportion of pig MAH isolates are imported from abroad. Importantly, we identified a novel lineage of MAH that was distributed in a specific area in Japan. Epidemiological survey and its unique genomic features allowed us to trace putative transmission routes via pig movement. Our study provides critical information for understanding the diversity of MAH population and the possibility of species-specificity in MAH. Our study will be useful for countermeasures of pig NTM disease.

    Staphylococcal enterotoxins (SEs) produced by enterotoxigenic Staphylococcus aureus in food cause staphylococcal food poisoning. We recently reported a foodborne outbreak due to S. aureus harboring new SE/SE-like (SEl) genes (seg, sei, sem, sen, seo, and selu) related to enterotoxin gene cluster (egc) 2 as with other research groups. However, the pathogenicity of SEs production remains unclear. Therefore, we herein investigated egc2-related SEs production from S. aureus isolates and leftover food items during a foodborne outbreak using a sandwich enzyme-linked immunosorbent assay suitable for the quantification of SEs. S. aureus isolates produced markedly high levels of egc2-related SEs, and the leftover food item "Sushi" contained SEs over the toxin dose that causes food poisoning symptoms. A representative isolate was subjected to whole-genome sequencing. The isolate was homologous with previously reported ST45 strains, particularly the unique genomic island νSaβ structure mostly consisting of egc2. The present study indicates that egc2-related SEs are food poisoning causative agents based on high SE production levels within an actual foodborne outbreak.

    Mycobacterium tuberculosis, the causative agent of tuberculosis, possess flavin-dependent thymidylate synthase, ThyX. Since thyX is absent in humans and was shown to be essential for M. tuberculosis normal growth, ThyX is thought to be an attractive novel TB drug target. In this study, we assessed thyX essentiality in Mycobacterium bovis BCG strains using CRISPR interference based gene silencing and found that thyX is not essential in an M. bovis BCG Tokyo derivative strain. We successfully constructed a thyX deletion mutant strain from that strain, which reinforces the non-essentiality of thyX under a certain genetic background. This article is protected by copyright.     <title>Abstract</title><italic>Mycobacterium avium</italic> subsp. <italic>hominissuis</italic> (MAH) is one of the most important agents causing non-tuberculosis mycobacterial infection in humans and pigs. Genome analysis on MAH of human isolates has been proceeding, however, those of pigs are limited despite its potential source of infection to human. In the current study, we obtained 30 draft genome sequences of MAH of pigs reared in Japan. The 30 draft genomes consisted of 4,848,678 – 5,620,788 bp length, 4,652 – 5,388 coding genes and 46 – 75 (Med: 47) tRNAs. All isolates had restriction modification associated genes and 185 – 222 predicted virulence genes. Two isolates had tRNA arrays and one isolate had a clustered regularly interspaced short palindromic repeat (CRISPR) region. Our results will be useful for evaluation of the ecology of MAH by providing a foundation for genome-based epidemiological studies.

    Rats are an important maintenance host of Leptospira spp., the causative agents of leptospirosis, a zoonotic disease with worldwide distribution. Rats are traded as food in the Mekong Delta, where Cambodia exports tons of rats to Vietnam. Handling wild rats is a potential health risk, but the information on Leptospira spp. carried by rats traded in the region remains limited. In this study, we investigated the carriage of Leptospira spp. in rats exported from Cambodia to Vietnam using bacterial culture, nested PCR and DNA sequencing. Isolates were then assessed using serological analysis and whole-genome sequencing (WGS), and Leptospira DNA detected in rat kidney tissues was also analysed by multilocus sequence typing (MLST). Seventy-two rats (2 Bandicota indica, 57 Rattus argentiventer, 11 R. losea, 1 R. norvegicus and 1 R. rattus) were subjected to bacterial culture, and three L. borgpetersenii serogroup Javanica isolates were obtained from R. argentiventer (5.3%). WGS revealed that although Cambodian isolates were genetically related to L. borgpetersenii serogroup Javanica strains widely distributed in East and South-East Asian countries, they formed a different cluster from other strains. In addition to the three L. borgpetersenii sequences, the flaB sequence of L. interrogans was detected in 18 R. argentiventer and 7 R. losea kidney tissue samples (38.9%) using nested PCR followed by DNA sequencing. The L. interrogans flaB-positive samples were further analysed by MLST, revealing that seven housekeeping genes (glmU, pntA, sucA, tpiA, pfkB, mreA and caiB) contained novel sequences with distinct lineages from other sequence types. This study revealed a high prevalence of Leptospira spp. among rats exported from Cambodia to Vietnam, indicating a potential risk to people engaging in rat trade and demonstrating that a fastidious L. interrogans strain circulates among Cambodian rats.

    <title>Abstract</title><sec>
    <title>Background</title>
    Tibetan highlanders have adapted to hypoxic environments through the development of unique mechanisms that suppress an increase in hemoglobin (Hb) concentration even in high-altitude areas. Hb concentrations generally decrease with increasing age. However, in the highlands, chronic altitude sickness is known to occur in the elderly population. To investigate how aging in a hypoxic environment affects Hb levels in Tibetan highlanders, we focused on the Mustang people, who live above 3500 m. We tried to clarify the pure relationship between aging and Hb levels in a hypoxic environment.
    </sec><sec>
    <title>Results</title>
    We found that the Hb concentration increased with increasing age in females but not in males. Multivariate analysis showed that age, pulse pressure, the poverty index, and vascular diameter were strongly correlated with the Hb concentration.
    </sec><sec>
    <title>Conclusions</title>
    We found unique Hb dynamics among female Tibetan highlanders. As seen in these Hb dynamics, there may be sex-based differences in the adaptive mechanism in Tibetan highlanders.

    Enterotoxigenic Escherichia coli (ETEC) strains that express various fimbrial or nonfimbrial colonization factors and enterotoxins are critical causes of diarrheal diseases. Human ETEC serotype O169:H41 (O169) has been a representative of epidemic ETEC worldwide; the organism shows massive adherence to HEp-2 cells similar to enteroaggregative E. coli. Previously, we determined the complete sequence of the unstable virulence plasmid, pEntYN10. The plasmid included a unique set of genes encoding a novel colonization factor (CF) resembling K88 (F4) of porcine ETEC, in addition to CS6, a well-known representative CF of human ETEC, and another novel CF similar to CS8 (CFA/III) of human ETEC. In the present study, we focused on K88-like CF (hereafter, K88O169) that may allow the organisms to infect domestic livestock like original K88-harboring strains that can cause diarrhea in piglets. Samples were tested for antibodies against recombinant proteins of possible paralogous adhesins, FaeG1 and FaeG2, from K88O169 and the FaeG of typical K88 (F4). The seroepidemiological study using recombinant antigens (two paralogs FaeG1 and FaeG2 from K88Oi69) showed reactivity of porcine (18.0%) and bovine (17.1%) sera to K88O169 FaeG1 and/or FaeG2 antigens on indirect ELISA tests. These results suggest that E. coli with K88O169 adhesin can infect various hosts, including pigs and cattle. This is the first report of domestic livestock having antibodies to K88O169 of human ETEC. Although human ETEC had been thought to be distinguished from those of domestic animals based on colonization factors, zoonotic strains may conceal themselves among human ETEC organisms.     <title>Abstract</title>
    While large scale mobility data has become a popular tool to monitor the mobility patterns during the COVID-19 pandemic, the impacts of non-compulsory measures in Tokyo, Japan on human mobility patterns has been under-studied. Here, we analyze the temporal changes in human mobility behavior, social contact rates, and their correlations with the transmissibility of COVID-19, using mobility data collected from more than 200K anonymized mobile phone users in Tokyo. The analysis concludes that by April 15th (1 week into state of emergency), human mobility behavior decreased by around 50%, resulting in a 70% reduction of social contacts in Tokyo, showing the strong relationships with non-compulsory measures. Furthermore, the reduction in data-driven human mobility metrics showed correlation with the decrease in estimated effective reproduction number of COVID-19 in Tokyo. Such empirical insights could inform policy makers on deciding sufficient levels of mobility reduction to contain the disease.

    <title>Abstract</title>
    <sec>
    <title>Background</title>
    Treatment of VRE is of clinical concern. While certain numbers of vanD-type VRE have been isolated, only two vanD5-harbouring Enterococcus faecium isolates have been reported in Canada and Japan.
    </sec>
    <sec>
    <title>Methods</title>
    We report the isolation of vanD5-type E. faecium and the first ever determination of the whole-genome sequence to investigate the possible mechanisms of the acquisition of the vanD5 gene cluster in E. faecium.
    </sec>
    <sec>
    <title>Results</title>
    Two vanD5-harbouring vancomycin-resistant E. faecium were isolated from the skin (SMVRE19) and faeces (SMVRE20) of a patient with a skin ulcer in Japan. The isolates exhibited vancomycin and teicoplanin MIC values of 128 mg/L, whilst the previous isolates of vanD5-harbouring E. faecium were only resistant to vancomycin. SMVRE19 and SMVRE20 were clones related to ST18, which is also seen in vanA- and vanB-type VRE. These isolates harboured an insertion element, ISEfm1, in the ddl gene, similar to a previously described teicoplanin-resistant vanD3-type E. faecium. The vanD5 gene cluster was integrated into the SMVRE20 chromosome as a part of a large genomic island (approximately 127 kb), similar to other recently spreading vanD variants in the Netherlands. The genomic island shared the greatest similarity with a part of the Blautia coccoides genome sequence, except for the region surrounding the vanD gene cluster.
    </sec>
    <sec>
    <title>Conclusions</title>
    This study reports that emergence of vancomycin- and teicoplanin-resistant vanD5-type E. faecium occurred via acquisition of the vanD5 cluster and ISEfm1 insertion into ddl. Considering the genetic similarity between the various VRE strains, the current study should serve as a warning against the spread of vanD5-type VRE.

    Nontuberculous mycobacteria (NTM) are opportunistic pathogens of humans and animals and are transmitted among the environment, wildlife, livestock, and humans. The aim of this study was to investigate the rate of isolation and antimicrobial susceptibility of NTM in wildlife. In total, 178 samples of feces (n=131) and tissues (n=47) were collected from 11 wildlife species in Gifu Prefecture and Mie Prefecture, Japan, between June 2016 and October 2018. We isolated NTM from 15.3% (20/131) of fecal samples using Ogawa medium, and isolates were identified by sequencing the rpoB and hsp65 genes. The rpoB sequences were compared with those from other strains of human and environmental origin. The NTM isolates were obtained from sika deer (Cervus nippon), wild boar (Sus scrofa), Japanese monkey (Macaca fuscata), raccoon dog (Nyctereutes procyonoides), masked palm civet (Paguma larvata), and Japanese weasel (Mustela itatsi) and were classified as rapidly growing mycobacteria (RGM) and slowly growing mycobacteria (SGM). The 12 RGM identified were Mycolicibacterium peregrinum (n=5), Mycolicibacterium fortuitum (n=3), Mycolicibacterium septicum (n=3), and Mycolicibacterium thermoresistibile (n=1), and the eight SGM were Mycobacterium paraense (n=4), Mycolicibacter arupensis (n=2), Mycolicibacter virginiensis (n=1), and Mycobacterium nebraskense (n=1). The NTM from wildlife showed ≥99% similarity with strains from different sources including humans. The RGM were susceptible to the antimicrobial agents tested except for M. fortuitum, which was resistant to azithromycin and clarithromycin. The SGM showed multiple drug resistance qualities but were susceptible to amikacin, clarithromycin, and rifabutin. These results indicate that wildlife may be reservoir hosts of NTM in Japan. The presence of antimicrobial-resistant NTM in wildlife suggests that the trends of NTM antimicrobial susceptibility in wildlife should be monitored.

    ネパールのムスタン群において、3570m高地に居住するツァラン村の住民を対象とし、非感染性疾患(NCD)の有病率と危険因子について横断調査を実施した。18歳以上の住民から無作為に選択した被験者188例に対しアンケート調査を行い、糖化ヘモグロビン(HbA1c)測定を含む生化学的検査および理学的検査を施行した。その結果、中程度の高血糖症および糖尿病の有病率はそれぞれ31.6%および4.6%で、血中酸素飽和度(SpO2)が90%未満である低酸素血症の有病率は27.1%であった。また、多重ロジスティック回帰分析からは、HbA1c≧6%の耐糖能異常有病率に関連する因子として、高年齢とSpO2低下であることが明らかにされた。以上より、ネパール高地人ではグルコース代謝障害で有病率が高く、要因として、高地生活の低酸素条件下による低酸素血症に関与するものと考えられた。

    In many countries with low to moderate tuberculosis (TB) incidence, cases have shifted to elderly persons. It is unclear, however, whether these cases are associated with recent Mycobacterium tuberculosis transmission or represent reactivation of past disease. During 2009-2015, we performed a population-based TB investigation in Yamagata Prefecture, Japan, using in-depth contact tracing and 24-loci variable-number tandem-repeat typing optimized for Beijing family M. tuberculosis strains. We analyzed 494 strains, of which 387 (78.3%) were derived from elderly patients. Recent transmission with an epidemiologic link was confirmed in 22 clusters (70 cases). In 17 (77.3%) clusters, the source patient was elderly; 11(64.7%) of the 17 clusters occurred in a hospital or nursing home. In this setting, the increase in TB cases was associated with M. tuberculosis transmissions from elderly persons. Prevention of transmission in places where elderly persons gather will be an effective strategy for decreasing TB incidence among predominantly elderly populations.

  • Mycolactone-producing Mycobacterium marinum infection in captive Hong Kong warty newts and pathological evidence of impaired host immune function

    A mass mortality event of captive Hong Kong warty newts Paramesotriton hong kongensis with non-granulomatous necrotic lesions occurred in Taipei Zoo, Taiwan, in 2014. Clinically, the sick newts were lethargic and often covered with water mold Saprolegnia sp. on the skin of the body trunk or extremities. Predominant pathological findings were multifocal non-granulomatous necrotic lesions in the liver, spleen, and kidneys and severe skin infection with Sapro legnia sp., with deep invasion and involvement of underlying muscles. The possibility of ranavirus infection was ruled out by negative PCR results. Unexpectedly, abundant intralesional acid-fast positive bacilli were found in the necrotic lesions of the liver, spleen, and kidney in all 14 sick newts. PCR targeting the hsp65, ITS region, and partial 16S rRNA genes was performed, and the sequence identity from amplified amplicons of hsp65 and partial 16S rRNA genes was 100% identical to that of the corresponding gene fragment of Mycobacterium marinum. Further molecular investigations demonstrated that the current M. marinum was a mycolactone-producing myco bacterium with the presence of esxA/esxB genes. Mycolactone is a plasmid-encoded, immunosuppressive, and cytotoxic toxin. The possible immunosuppression phenomenon characterized by systemic non-granulomatous necrotic lesions caused by M. marinum and the unusual deep invasive infection caused by water mold might be associated with the immunosuppressive effect of mycolactone. Therefore, it should be noted that non-granulomatous necrotic lesions in amphibians can be caused not only by ranavirus infection but also by mycobacteriosis.

    Japan Anti-Tuberculosis Association(12)-VNTR[JATA(12)-VNTR]結核菌型別解析法のプロトコルに基づいて、2014年4月から12月に新規登録された結核患者から分離された24菌株の型別を調査し、既に報告した103菌株と合わせた127菌株についてクラスター分類を行った。今回解析した24菌株のうち5菌株が関連するクラスターは1つ、2菌株が関連するクラスターは3つ、1菌株が関連するクラスターは5つの合計9クラスターに分類され、16菌株がいずれかのクラスターに包含された。また、実地疫学調査では個別の感染経路については解明できなかったが、多くの菌株で構成されるクラスターは和田らが報告した阪神地区において出現頻度の高いJATA(12)-VNTR型別に一致し、京都府内でも同様の型別の流行があると予測される。

    2012年1月〜2014年12月の間の大阪市における新登録結核患者は3204名であった。今回、結核菌が分離搬入された1654株中、687株のJATA(12)-VNTR型別を実施した。その結果、1)687株のうち、ハイリスクグループはホームレス患者由来68株、外国出生患者28株、若年患者由来187株、多剤耐性結核菌(MDR)患者由来18株であった。2)JATA(12)-VNTR型別より12領域での反復数が一致したものをクラスターと定義したところ、419株(61.0%)が86クラスターを形成しており、クラスタータイプ別の出現割合はJ12-0001が40株(5.8%)と最も多く、2006〜2011年の出現割合とほぼ同様であった。3)M株を示すクラスタータイプのJ12-0006は11株検出され、ホームレス患者由来の68株の最も大きなクラスターは7株から形成されるJ12-0010 で、このうち6株は追加領域を含めた24領域のVNTR型が全て一致した。4)外国出生患者由来28株の14株が国内出生患者由来株と11クラスターを形成していたが、このうち5クラスターは追加領域のVNTR型が不一致であり、国内出生患者由来株とは異なる菌株であることが判明した。5)若年患者由来187株間では91株が30クラスターを形成し、最も大きなクラスターはJ12-0001とクラスターG各々9株であった。尚、MDR18株中3株はJ12-0022で、このタイプの菌株が出現した場合はMDRの可能性に注意が必要と考えられた。

    Japan Anti-Tuberculosis Association(12)-VNTR結核菌型別解析法において高分子量領域に対応できるアガロースゲル電気泳動法について検討し、バイオラッド製Sub-Cell GT電気泳動槽を用いて360分間の泳動を行うことで解析できることを確認した。本法を用いて2013年度に分離・保存された菌株43株の解析を行った結果、既報60菌株と併せ、10菌株、5菌株、4菌株、3菌株、2菌株で構成されるクラスターがそれぞれ1個、1個、3個、3個、8個と、クラスターを構成しない菌株に分類された。今回解析の43株に関連する10個のクラスター型について実地疫学調査を行ったところ、山城北保健所管内と丹後保健所管内の両方の菌株が含まれた5個のクラスターでは患者間の関連性を解明できなかったが、一方のみの菌株で構成された5個中2個のクラスターの事例では感染経路が解明され、結核対策に結びつけることができた。

    Four Japanese forest green tree frogs (Rhacophorus arboreus) were presented with emaciation, abdominal distention and ulcerative and nodular cutaneous lesions affecting the brisket, limbs, digits and ventral abdomen. Another three frogs had been found dead in the same tank 1 year previously. Necropsy examination of these seven frogs revealed splenomegaly and hepatomegaly, with multiple tan-yellow nodular foci present in the liver, spleen, heart, lungs, ovaries and kidneys. Microscopically, five frogs had necrosis and surrounding granulomatous inflammation in the liver, spleen, kidneys, lungs, intestine and ovaries, with numerous acid-fast bacilli in the areas of necrosis. Two frogs had granulomatous lesions in the lungs, liver, spleen, heart, coelomic membrane, stomach and intestinal wall. These lesions had no or minimal necrosis and few acid-fast bacilli. Mycobacterium spp. was cultured from three frogs and identified as Mycobacterium marinum by colony growth rate and photochromogenicity and DNA sequencing. This is the first report of M. marinum infection in Japanese forest green tree frogs. (C) 2014 Elsevier Ltd. All rights reserved.

    Capnocytophaga canimorsus, which is often found in the oral cavities of dogs and cats, is sometimes transmitted to humans, causing severe infection. To elucidate the risk of C canimorsus in humans and animals, this study was undertaken to characterize this bacterium epidemiologically and genetically. We examined the distribution of C canimorsus in dogs and cats, and analyzed the correlation between the presence of bacteria and individual factors statistiCally. We also compared C canimorsus isolates genetically using 16S rRNA gene sequence analysis and pulsed-field gel electrophoresis (PFGE). C canimorsus was detected in 76 of 109 dogs (69.7%) and 57 of 104 cats (54.8%). A relation between C canimorsus presence and some individual factors was detected both in dogs and cats, but the predictive factors of carrying the bacterium differed between dogs and cats. 16S rRNA gene sequences from C canimorsus isolates in this study were combined with previously published sequences to assess their intra-specific phylogeny. Results show that C. canimorsus is classifiable into two main groups (I and II) with differing -y-glutamyl aminopeptidase activity. Strains from human patients belonged unevenly to group I, possibility suggesting that group I can be transmitted to humans and group II is indigenous only to the oral cavities of dogs and cats. PFGE genotyping showed high discriminatory power, and the dendrogram accorded with genetic segregation between isolates of group land II. Sma I-digest PFGE developed for this study is useful as a molecular typing method for additional epidemiological and phylogenetic studies of C canimorsus. (C) 2014 Elsevier B.V. All rights reserved.

    BACKGROUND: The quality of variable number of tandem repeats (VNTR) typing of Mycobacterium tuberculosis was first investigated in 2009 in 37 laboratories worldwide. The results revealed an inter- and intra-laboratory reproducibility of respectively 60% and 72%. These data spurred an improvement in laboratory-specific assays and global standardisation of VNTR typing.
    OBJECTIVE: To measure the effects of the technical improvements and increased standardisation, a test panel consisting of 30 M. tuberculosis complex DNA samples was distributed for VNTR typing in 41 participating laboratories from 36 countries.
    RESULTS: The inter- and intra-laboratory reproducibility increased overall to respectively 78% and 88%. The 33 laboratories that participated in both the first and second proficiency studies improved their inter- and intra-laboratory reproducibility from 62% and 72% to respectively 79% and 88%. The largest improvement in reproducibility was detected in 10 laboratories that use an in-house polymerase chain reaction technique and perform amplicon sizing using gel electrophoresis. Detailed error analysis revealed a reduction in the number of systematic errors, sample exchange events and non-amplifiable loci.
    CONCLUSION: This second worldwide proficiency study indicates a substantial increase in the reproducibility of VNTR typing of M. tuberculosis. This will contribute to a more meaningful interpretation of molecular epidemiological and phylogenetic studies on the M. tuberculosis complex.

    大阪市内で食中毒が疑われ、当研究所に検体が搬入された事件についてまとめ、その原因病原体と疫学情報を基に感染源を明らかにした。食中毒が疑われた患者の便と吐物、食品残品、原因と考えられる施設(患者宅を含む)に保存されていた検食と同施設のふきとり水および調理従事者の便と手指のふきとり水を調査の対象とした。その結果、2012年に食中毒、有症苦情などで検体が搬入されたのは117事件の検体で、68件に下痢原性微生物が検出された。なお68件の内訳はノロウイルス47件(69.1%)、カンピロバクター7件(10.3%)、サルモネラ属菌5件(7.4%)、黄色ブドウ球菌、クドア・セプテンプンクタータ2件(2.9%)、毒素原性大腸菌0153、腸管出血性大腸菌026、セレウス菌、サポウイルスが各1件、そして黄色ブドウ球菌とカンピロバクターが同時に検出された事件も1件に認められた。68件のうち、推定原因食品が判明した事件数は15件(22.1%)であり、15件のうち検出された原因微生物の内訳はノロウイルスが10件、クドア2件、サルモネラ1件、黄色ブドウ球菌1件、セレウス菌1件であり、原因施設が特定された事件数は63件(92.6%)に認められ、推定された原因食品は生カキ、弁当、寿司、コース料理、スパイスソース、ヒラメ、カレー、刺身などで、原因施設は飲食店が例年通り最も多く認められた。大阪市における細菌あるいはウイルス性食中毒事件として厚生労働省に届けられた事件数は24件であったが、全国的にもノロウイルス、カンピロバクターの食中毒事件の占める割合が多く、今後も十分な注意が必要であり、消費者にはきちんと加熱して食する習慣を守ること、平常時を含めた二次感染防止策の徹底・予防の啓発が重要と考えられた。

    前田らが確立した結核菌分類法のJapan Anti-Tuberculosis Association(JATA) 12-VNTR(Variable Number of Tandem Repeats)を用い、2004年4月〜2012年3月に分離・保存した42菌株と2012年4月〜2013年3月に入手した18菌株の合計60菌株の型別解析を行った。その結果、60菌株のVNTR型は45のパターンに分類され、このうち24菌株で9個のクラスターを形成した。このクラスターごとに感染経路調査を実施したところ、9事例中3事例で解明され、うち1例は追跡調査に発展した。JAYA(12)-VNTR型別解析は、菌株間の関連が明白な場合や感染源・経路が実地疫学的に予想されている場合には極めて有用であり、これらの結果は結核対策の科学的裏付けや追跡調査に活用できると考えられた。

    2011年に大阪市内の食中毒原因調査において検出された下痢原性微生物について報告した。対象は119事例1123検体で、68件より下痢原性微生物が検出された。内訳はノロウイルス38件、カンピロバクター19件、サルモネラ属菌3件、黄色ブドウ球菌2件、ウェルシュ菌、毒素原性大腸菌O159、腸管出血性大腸菌O157、セレウス菌、サポウイルス各1件で、クドア・セプテンプンクタータ/カンピロバクター同時検出が1件であった。ノロウイルスの検出は12〜1月がピークで、2010年と同様にGenogroup IIが主流であったが、Genogroup Iも5件検出された。原因食品でカキ関連が明らかになっていたのは2件のみで、他の食品関連が例年同様多かった。カンピロバクターは2009年から連続して増加しており、原因食品は不明が多かった。全68件のうち推定原因食品が判明したのは37件、原因施設が特定されたのは45件であった。

    Beijing family strains of Mycobacterium tuberculosis have attracted worldwide attention because of their wide geographical distribution and global emergence. Peru, which has a historical relationship with East Asia, is considered to be a hotspot for Beijing family strains in South America. We aimed to unveil the genetic diversity and transmission characteristics of the Beijing strains in Peru. A total of 200 Beijing family strains were identified from 2140 M. tuberculosis isolates obtained in Lima, Peru, between December 2008 and January 2010. Of them, 198 strains were classified into sublineages, on the basis of 10 sets of single nucleotide polymorphisms (SNPs). They were also subjected to variable number tandem-repeat (VNTR) typing using an international standard set of 15 loci (15-MIRU-VNTR) plus 9 additional loci optimized for Beijing strains. An additional 70 Beijing family strains, isolated between 1999 and 2006 in Lima, were also analyzed in order to make a longitudinal comparison. The Beijing family was the third largest spoligotyping clade in Peru. Its population structure, by SNP typing, was characterized by a high frequency of Sequence Type 10 (ST10), which belongs to a modern subfamily of Beijing strains (178/198, 89.9%). Twelve strains belonged to the ancient subfamily (ST3 [n = 3], ST25 [n = 1], ST19 [n = 8]). Overall, the polymorphic information content for each of the 24 loci values was low. The 24 loci VNTR showed a high clustering rate (80.3%) and a high recent transmission index (RTIn-1 = 0.707). These strongly suggest the active and ongoing transmission of Beijing family strains in the survey area. Notably, 1 VNTR genotype was found to account for 43.9% of the strains. Comparisons with data from East Asia suggested the genotype emerged as a uniquely endemic clone in Peru. A longitudinal comparison revealed the genotype was present in Lima by 1999.

    The Beijing family is an endemic lineage of Mycobacterium tuberculosis in eastern Asia. In Japan, five evolutionarily sequential sublineages composing the lineage have predominated. Comparative genomic sequencing based on a microarray technique was conducted for five representative strains of those respective sublineages. Results revealed approximately 200 point mutations specific to each strain. Subsequently, to investigate the genetic diversity of each sublineage, we analysed the phylogenetic divergence of 103 domestic strains belonging to them using genetic markers derived from the mutation information. Results show that the five sublineages have comprised smaller lineages which had diverged at various points. The smaller sub-sublineages have emerged with respective bottlenecks, which are reflected in the excessive monophyletic evolution of the species. Our data provide necessary information to grasp a comprehensive picture of genetic diversity of the lineage constructed in its evolution. (c) 2012 Elsevier B.V. All rights reserved.

    Infection and transmission of multidrug-resistant Mycobacterium tuberculosis (MDR-Mtb) and extensively drug-resistant M. tuberculosis (XDR-Mtb) is a serious health problem. We analyzed a total of 1,110 Mtb isolates in Osaka Prefecture and neighboring areas from April 2000 to March 2009. A total of 89 MDR-Mtb were identified, 36 (48.5%) of which were determined to be XDR-Mtb. Among the 89 MDR-Mtb isolates, 24 (27.0%) phylogenetically distributed into six clusters based on mycobacterial interspersed repetitive units-various number of tandem repeats (MIRU-VNTR) typing. Among these six clusters, the MIRU-VNTR patterns of four (OM-V02, OM-V03, OM-V04, and OM-V06) were only found for MDR-Mtb. Further analysis revealed that all isolates belonging to OM-V02 and OM-V03, and two isolates from OM-V04 were clonal. Importantly such genotypes were not observed for drug-sensitive isolates. These suggest that few but transmissible clones can transmit after acquiring multidrug resistance and colonize even in a country with a developed, well-organized healthcare system.

  • First Worldwide Proficiency Study on Variable-Number Tandem-Repeat Typing of Mycobacterium tuberculosis Complex Strains

    Although variable-number tandem-repeat (VNTR) typing has gained recognition as the new standard for the DNA fingerprinting of Mycobacterium tuberculosis complex (MTBC) isolates, external quality control programs have not yet been developed. Therefore, we organized the first multicenter proficiency study on 24-locus VNTR typing. Sets of 30 DNAs of MTBC strains, including 10 duplicate DNA samples, were distributed among 37 participating laboratories in 30 different countries worldwide. Twenty-four laboratories used an in-house-adapted method with fragment sizing by gel electrophoresis or an automated DNA analyzer, nine laboratories used a commercially available kit, and four laboratories used other methods. The intra-and interlaboratory reproducibilities of VNTR typing varied from 0% to 100%, with averages of 72% and 60%, respectively. Twenty of the 37 laboratories failed to amplify particular VNTR loci; if these missing results were ignored, the number of laboratories with 100% interlaboratory reproducibility increased from 1 to 5. The average interlaboratory reproducibility of VNTR typing using a commercial kit was better (88%) than that of in-house-adapted methods using a DNA analyzer (70%) or gel electrophoresis (50%). Eleven laboratories using in-house-adapted manual typing or automated typing scored inter-and intralaboratory reproducibilities of 80% or higher, which suggests that these approaches can be used in a reliable way. In conclusion, this first multicenter study has documented the worldwide quality of VNTR typing of MTBC strains and highlights the importance of international quality control to improve genotyping in the future.

    2010年に食中毒、有症苦情などで検体が搬入された122事件1,624検体について、下痢原性微生物の検出を行った。122事件中73件から下痢原性微生物が検出され、大阪市における細菌あるいはウイルス性食中毒事件として厚生労働省に届けられたのは26件であった。また、73件から検出された病原体の内訳はノロウイルス47件、カンピロバクター14件、サルモネラ属菌4件、下痢原性大腸菌3件、ウェルシュ菌2件と、黄色ブドウ球菌、腸炎ビブリオ、黄色ブドウ球菌/ウェルシュ菌同時検出が各1件であった。2010年は2002年以降の傾向と同様にノロウイルス、カンピロバクター、サルモネラによる食中毒が多く発生したが、特にノロウイルスによる事件の多さが際立っていた。

    2009年に市内で食中毒が疑われ当研究所に検体が搬入された事件数は115件で、このうち下痢原生微生物が検出されたのは62件(54%)であった。検出微生物の内訳はノロウイルス41件(66%)、カンピロバクター10件(16%)、サルモネラ属菌6件(10%)、黄色ブドウ球菌3件(5%)、毒素原生大腸菌2件(3%)、ウェルシュ菌1件(1.6%)、O157 1件(1.6%)で、ノロウイルスとカンピロバクターの同時検出例が2件(3%)あった。62件のうち推定原因食品が判明したのは26件(42%)で、主な原因食品は「コース料理」「会席料理」「鶏料理」などであった。原因施設は飲食店が多かった。

    A population-based study of Mycobacterium tuberculosis isolated from homeless tuberculosis patients was performed during 2002-2004 in Osaka City, Japan. The data show that the ancient Beijing subfamily was predominant, whereas clustered isolates based on refined variable number of tandem repeats genotyping (19 loci) mainly belonged to the modern Beijing subfamily, suggesting its increased transmissibility.

    2007年に大阪市内の食中毒原因調査において検出された下痢原性微生物について報告した。食中毒が疑われた患者の便と吐物、食品残品、原因と考えられる施設(患者宅を含む)に保存されていた検食と同施設のふきとり水および調理従事者の便と手指のふきとり水を調査の対象とした。検体が搬入された事件数は合計139件で、患者および関係者の検便および吐物1482検体、食品、ふき取りなどが892検体であった。139事件中66件から下痢原性微生物を検出した。139件のうち大阪市における細菌あるいはウイルス性食中毒事件として厚生労働省に届けられた事件数は33件であった。下痢原性微生物が検出された66事件の内訳は、ノロウイルスが29件で最も多く、以下、カンピロバクター19件、サルモネラ11件、黄色ブドウ球菌4件、腸炎ビブリオ2件、ウェルシュ菌1件であった。

    大阪市内で食中毒が疑われ検体が搬入された事件について毎年まとめ、その原因病原体と疫学情報を基に感染源を明らかにした。2006年に大阪市内で食中毒が疑われた202事件について下痢原性微生物の検出を行った。下痢原性微生物が検出された121事件(59.9%)のうち、ノロウイルスが検出されたものが94件(77.7%)、カンピロバクター12件(9.9%)、サルモネラ7件(5.8%)、腸炎ビブリオ3件(2.5%)、セレウス菌2件、黄色ブドウ球菌1件、病原大腸菌1件、エロモナス1件であった。また、市内の一ホテルにおいて患者数160例のノロウイルスによる大規模食中毒事件が発生した。

    大阪市内で分離した結核菌の分子疫学的解析,薬剤耐性結核の出現に対する危機管理に関して,従来法に代わる新手法についてその有用性を検討した.結核菌株251株についてMIRU-VNTR法による分子疫学的解析を行った.MIRU-VNTR法は患者接触調査などと合わせた感染源調査では充分な威力を発揮することが予想された.ただし,MIRU-VNTRでの分離が難しいパターンの存在が明らかになった.対照プローブと他のプローブの蛍光強度を比較することによって,目的遺伝子内の変異を検出できることが明らかとなった.さらに,これらのプローブ濃度を調整することで,一定の蛍光強度に至るために必要なPCRサイクル数を比較するだけで突然変異の有無が判定できるシステムとして完成させた

    Mutations in particular nucleotides of genes coding for drug targets or drug-converting enzymes lead to drug resistance in Mycobacterium tuberculosis. For rapid detection of drug-resistant M. tuberculosis in clinical specimens, a simple and applicable method is needed. Eight TaqMan minor groove binder (MGB) probes, which discriminate one-base mismatches, were designed (dual-probe assay with four reaction tubes). The target of six MGB probes was the rpoB gene, which is involved in rifampin resistance; five probes were designed to detect for mutation sites within an 81-bp hot spot of the rpoB gene, and one probe was designed as a tuberculosis (TB) control outside the rpoB gene hot-spot. We also designed probes to examine codon 315 of katG and codon 306 of embB for mutations associated with resistance to isoniazid and ethambutol, respectively. Our system was M. tuberculosis complex specific, because neither nontuberculous mycobacteria nor bacteria other than mycobacteria reacted with the system. Detection limits in direct and preamplified analyses were 250 and 10 fg of genomic DNA, respectively. The system could detect mutations of the rpoB, katG, and embB genes in DNAs extracted from 45 laboratory strains and from sputum samples of 27 patients with pulmonary TB. This system was much faster (3 h from DNA preparation) than conventional drug susceptibility testing (3 weeks). Results from the dual-MGB-probe assay were consistent with DNA sequencing. Because the dual-probe assay system is simple, rapid, and accurate, it can be applied to detect drug-resistant M. tuberculosis in clinical laboratories.

    Mycobacterium tuberculosis infects not only host macrophages but also nonprofessional phagocytes, such as alveolar epithelial cells. Glycosaminoglycans (GAGs) are considered as the component of mycobacterial adherence to epithelial cells. Here we show that extracellularly occurring mycobacterial DNA-binding protein 1 (MDP1) promotes mycobacterial infection to A549 human lung epithelial cells through hyaluronic acid ( HA). Both surface plasmon resonance analysis and enzyme-linked immunosorbent assay revealed that MDP1 bound to HA, heparin, and chondroitin sulfate. Utilizing synthetic peptides, we next defined heparin-binding site of 20 amino acids from 31 to 50 of MDP1, which is responsible for the specific DNA-binding site of MDP1. MDP1 bound to A549 cells, and exogenous DNA and HA interfered with the interaction. The binding was also abolished by treatment of A549 cells with hyaluronidase, suggesting that HA participates in the MDP1-A549 cell interaction. Adherence of bacillus Calmette-Guerin ( BCG) and M. tuberculosis to A549 cells was inhibited by addition of HA, DNA, and anti-MDP1 antibody, showing that MDP1 participates in the interaction between mycobacteria-alveolar epithelial cells. Simultaneous treatment of intratracheal BCG-infected mice with HA reduced the growth of BCG in vivo. Taken together, theses results suggest that HA participates in Mycobacterium-lung epithelium interaction and has potential for therapeutic and prophylactic interventions in mycobacterial infection.

    Diarrheagenic Escherichia coli (DEC) represents an elusive target, since they are not easily distinguished from fecal coliforms. To clarify if DEC are prevalent among sporadic cases of diarrheal illness in Osaka City Japan, diarrheal specimens were examined for E. coli that were enterohemorrhagic (EHEC), enteiopathogenic (EPEC), enterotoxigenic (ETEC), enteroinvasive (EIEC), or enteroaggregative (EAggEC). EAST1EC, a strain of E. coli that does not possess any diarrheagenic characteristics except the EAggEC heat-stable toxin I (EAST1) gene, was also included as a possible DEC. A total of 924 specimens were examined between July 1997 and March 2000. DEC and Salmonella were isolated from 7.3% (67/924) and 6.8% (63/924) of specimens, respectively. DEC was therefore as prevalent as Salmonella among sporadic cases. The 67 strains were composed of 17 EPEC (26%), 10 EHEC (15%), four ETEC (6%), 13 EAggEC (20%), and 23 EAST1EC (35%), including two. strains of EAST1EC 0 166:H15. Although PCR and tissue culture adhesion tests were useful to detect DEC, the effectiveness of serotyping was limited: only 40 strains (17.5%) out of 229 isolates that had been assumed to be enterovirulent on the basis of their 0 antigen were recognized to be diarrheagenic. In conclusion, not only EHEC but also the other subgroups of DEC, including EAST1EC, seem to play an important role, in causing sporadic diarrheal illnesses. Methods to detect and unified criteria to identify various kinds of DEC are strongly desired.

    HSP47 is a collagen-specific molecular chaperone that specifically recognizes and binds to the triple helical domain of various types of collagens. Here we report the cloning of the entire coding region of a novel collagen-like protein by yeast two-hybrid screening of a 17.5-day whole mouse embryo cDNA library using HSP47 as a bait. The cDNA of this protein and its deduced amino acid sequence are 2,690 bp and 438 amino acids long, respectively. The protein contains two clusters of Gly-X-Y collagenous repeats and three noncollagenous domains. Northern blot analysis showed that its mRNA is specifically expressed in the testis and ovary in adult tissues and that expression in these tissues is highest in the neonate. Biochemical characterization of this protein revealed that its proline residues are hydroxylated, it undergoes N-linked glycosylation, it forms trimers, and it is secreted in vitro. Immunohistochemical studies showed that the myoid cells and the pre-theca cells synthesized it in the testis and ovary, respectively, resulting in the accumulation of this protein in the extracellular spaces of these organs. these observations suggest that this protein is a new member of the collagen protein family. We thus designated this protein as type XXVI collagen.
