    The dinuclear oxidovanadium(IV/V) complex, [V2(HS2O8)O3(C18H24N2)2] - CH3CN or [V2O2(μ-O)( μ-H(SO4)2)(4,4'-tBubpy)2] CH3CN (4,4'-tBubpy = 4,4'- di-tert-butyl-2,20-bipyridine), has crystallographic C2 symmetry and exhibits a distorted octahedral geometry around the vanadium center, where the two 4,4'- tBubpy ligands are nearly orthogonal to each other. The two vanadium ions are linked by an oxo anion and a unique protonated sulfate anion [H(SO4)2 3- ]. In the crystal, intermolecular C-H πand π- π interactions between the 4,4'- tBubpy ligands are present, leading to a three-dimensional network.

    BACKGROUND: Photodynamic therapy (PDT) using photosensitisers is a minimally invasive treatment for malignant tumours. However, ideal photosensitisers are not yet established. Recently, we developed a new photosensitiser, glucose-conjugated chlorin e6 (G-Ce6). OBJECTIVES: To evaluate the clinical efficacy of vascular-targeted PDT (VTP), a type of PDT utilising a short drug-light interval, using G-Ce6 to treat spontaneously occurring tumours in dogs. METHODS: Five dogs with spontaneously occurring tumours (malignant melanoma: three; haemangiopericytoma: two; and squamous cell carcinoma: one) were subjected to VTP. These dogs were intravenously injected with G-Ce6 at doses of 1-3 mg/kg 5 min before laser irradiation. Tumours were superficially or interstitially irradiated using a 677-nm diode laser. RESULTS: Repeated VTP decreased tumour size, yielding complete remission in three dogs. Complications such as oedema surrounding normal tissues and fistulae were observed, and the oedema was self-limiting. The fistula was cured by debriding the necrotic tissues formed after VTP. CONCLUSIONS: Our findings demonstrate that VTP using G-Ce6 had antitumour effects in dogs with spontaneously occurring tumours.

    <p>A long-standing goal in the field of cancer therapy is the ability to treat tumors noninvasively. Photodynamic therapy (PDT) has attracted attention as a less invasive method for treating cancer. Thus, we focused on the sugar conjugation of photosensitizers (chlorin, C<sub>60</sub> etc.), mainly to improve their biocompatibility and tumor selectivity. We have developed sugar (glucose, mannose, maltotriose etc.)-conjugated chlorins as third-generation photosensitizers for PDT and glucose conjugated chlorin e6 (G-Ce6), especially, shows excellent PDT effects <i>in vitro</i> and <i>in vivo</i>. In addition, excellent PDT effects have been observed in dog cancer models.</p>

    Nanosheets of metalorganic frameworks (MOFs)®porous crystalline materials consisting of metal ions and organic ligands®are actively studied for their intrinsic chemical/physical properties attributed to the reduced dimensionality and for their potential to function as ideal components of nanodevices, especially when electrical conduction is present. Air/liquid interfacial synthesis is a promising technique to obtain highly oriented MOF nanosheets. However, rational control of size and shape combined with the aimed functionality remains an important issue to address making it necessary to research the critical factors governing nanosheet characteristics in the interfacial synthesis. Here, we investigate the influence of the solvent®methanol (MeOH) versus N, N-dimethylformamide (DMF)®used to prepare a ligand spread solution on an assembly of MOF nanosheets composed of Ni2+ and 2,3,6,7,10,11-hexaiminotriphenylene (HITP) (HITP-Ni-NS). We find that the macroscopic morphological uniformity in the micrometer scale is higher when DMF is used as the solvent. Regarding the microscopic crystalline domain, molecules of DMF with relatively high polarity and boiling point are involved in HITP-Ni-NS formation, hindering its growth and resulting in nanosheets with slightly smaller lateral size than that grown when MeOH is used. These findings provide crucial guidelines towards establishing a judicious strategy for creating desired MOF nanosheets at the air/liquid interface, thereby driving forward research on both fundamental and applied aspects of this field.

    <p>M2-like tumor-associated macrophages (M2-TAMs) in cancer tissues play a key role in cancer progression and immune evasion. Therefore, cancer immunotherapies targeting M2-TAMs have attracted considerable attention. Photodynamic therapy (PDT) is a minimally invasive and site-selective approach that kills target cells using a visible laser and a nontoxic photosensitizer. The photosensitizer is activated by the laser and produces reactive oxygen species (ROS) that induce cell death of target cells. We developed a novel photosensitizer, mannose-conjugated chlorin e6 (M-chlorin e6), which can be selectively delivered to M2-TAMs that highly express mannose receptors (CD206). We evaluated the anti-tumor effect of M-chlorin e6 PDT in a mouse model of allogeneic transplantation of CT26 cells, a murine colon cancer cell line. M-chlorin e6 PDT significantly reduced tumor volume and weight compared to the control group. M-chlorin e6 PDT also decreased the percentage of M2-TAMs (CD11b<sup>+</sup>, CD206<sup>+</sup>) and increased the percentage of M1-TAMs (CD11b<sup>+</sup>, CD80<sup>+</sup> or CD86<sup>+</sup>, CD206<sup>-</sup>) in the tumor tissue. Moreover, M-chlorin e6 PDT directly inhibited cancer cell viability and enhanced phagocytosis by the macrophage cell line RAW264.7. These results suggest that M-chlorin e6 PDT exerts its anti-tumor effect by reduction of M2-TAMs, inducing cancer cell death, and promoting macrophage phagocytosis.</p>

    Photodynamic therapy (PDT) damages cancer cells via photosensitization using harmless laser irradiation. We synthesized a new photosensitizer, mannose-conjugated-chlorin e6 (M-chlorin e6), which targets mannose receptors that are highly expressed on M2-like tumor-associated macrophages (M2-TAMs) and cancer cells. In our previous study, we demonstrated that M-chlorin e6 PDT reduces tumor volume and decreases the proportion of M2-TAMs. Whether M-chlorin e6 PDT-treated cancer cells activate tumor immunity remains unclear, although the decrease in M2-TAMs is thought to be a direct injurious effect of M-chlorin e6 PDT. Calreticulin (CRT) is exposed at the surface of the membrane of cancer cells in response to treatment with chemotherapeutic agents such as anthracycline and oxaliplatin. Surface-exposed CRT induces phagocytosis of CRT receptor-positive cells, including macrophages, inducing anticancer immune responses. In the present study, we found that M-chlorin e6 PDT increases CRT on the surface of cancer cells, leading to macrophage phagocytosis of cancer cells. Furthermore, M-chlorin e6 PDT increases CD80+CD86+ macrophages. These results suggest that M-chlorin e6 PDT exerts anti-tumor effects by both enhancing the phagocytosis of cancer cells and strengthening the anti-tumor phenotype of macrophages.

    A sustainable advanced synthetic method was developed based on redox cyclization for the synthesis of 2-arylbenzothiazoles in good to excellent yields from readily available nitrobenzenes, benzylamines, and elemental sulfur without the use of transition-metal catalysts. This method is remarkable: nitro group reduction, a benzylamine redox reaction, sulfuration, condensation, and cyclization, all proceed in a single step to generate a heterocycle. It is also highly atom-economical and does not require any external oxidizing or reducing agents.

    Conventional quinazoline synthesis methods involve a highly multistep reaction, and often require excess amounts of substrate to control the product selectivity, leading to significant resource wastage. Hence, in this study, from the viewpoint of green chemistry, we developed a novel metal-free synthetic method for 2-substituted quinazoline derivatives by the 4,6-dihydroxysalicylic acid-catalyzed oxidative condensation of <italic>o</italic>-aminobenzylamines and benzylamines using atmospheric oxygen. In this system, the use of a catalytic amount of BF<sub>3</sub>‧Et<sub>2</sub>O (10 mol%) as a Lewis acid successfully led to the efficient oxidative condensation and intramolecular cyclization of these amines, followed by aromatization to afford the corresponding 2-arylquinazolines in up to 81% yield with excellent atom economy and environmental factor. Furthermore, to expand this green oxidation method to gram-scale synthesis, we investigated the development of an oxidation process using salicylic acid itself as an organocatalyst, and established a method for the practical green synthesis of a series of nitrogen-containing heterocycles. We expect that the findings will contribute to the development of practical synthesis methods for pharmaceutical manufacturing and industrial applications, along with further advancements in green chemistry.

    The addition of interelement compounds with heteroatom-heteroatom single bonds to carbon-carbon unsaturated bonds under light irradiation is believed to be an atomically efficient method to procure materials with carbon-heteroatom bonds. In this study, we achieved the photoin-duced bisphosphination of alkynes using the phosphorus interelement compound, tetraphenyldiphos-phine monosulfide (1), to stereoselectively obtain the corresponding (E)-vic-1,2-bisphosphinoalkenes, which are important transition-metal ligands. The bisphosphination reaction was performed by mixing 1 and various alkynes and then exposing the mixture to light irradiation. Optimization of the conditions for the bisphosphination reaction resulted in a wide substrate range and excellent trans-selectivity. Moreover, the completely regioselective introduction of pentavalent and trivalent phosphorus groups to the terminal and internal positions of the alkynes, respectively, was achieved. We also found that the novel double-bond isomerization reaction of the synthesized bisphosphinated products occurred with a catalytic amount of a base under mild conditions. Our method for the pho-toinduced bisphosphination of carbon-carbon unsaturated compounds may have strong implications for both organic synthesis and organometallic and catalyst chemistry.

    Although diaryl tellurides are parent organotellurium compounds, their synthesis methods, especially for unsymmetrical ones, are limited. This may be due to the instability of diaryl tellurides and their synthesis intermediates under reaction conditions. Radical reactions are known to exhibit excellent functional group selectivity; therefore, we focused on a bimolecular homolytic substitution (SH2) reaction between the aryl radical and diaryl ditelluride. Aryl radicals are generated from arylhydrazines in air and captured by diaryl ditellurides, resulting in a selective formation of unsymmetrical diaryl tellurides with high yields. The electronic effects of the substituents on both arylhydrazines and diaryl ditellurides on the SH2 reaction of tellurium are also discussed in detail.

    Endohedral metal hemicryptophanes are attractive metal complexes with secondary coordination sphere models. Here, an endohedral Zn(II) hemicryptophane complex is directly synthesized using a triethylene glycol chain as a moderate length spacer between the cyclotriguaiacylene (CTG) and quinolinol units of the novel tripodal ligand. The guest of choline was encapsulated within the cavity of Zn(II) hemicryptophane complex.

    A simple and efficient method for α-brominating lactones that affords α<italic>-</italic>bromolactones under mild conditions using tetraalkylammonium hydroxide (R<sub>4</sub>N<sup>+</sup>OH<sup>−</sup>) as a base was developed. Lactones are ring-opened with Br<sub>2</sub> and a substoichiometric amount of PBr<sub>3</sub>, leading to good yields of the corresponding α-bromocarboxylic acids. Subsequent intramolecular cyclization over 1 h using a two-phase system (H<sub>2</sub>O/CHCl<sub>3</sub>) containing R<sub>4</sub>N<sup>+</sup>OH<sup>−</sup> afforded α-bromo lactones in good yields. This method can be applied at the 10 mmol scale using simple operations. α-Bromo-δ-valerolactone, which is extremely reactive and difficult to isolate, could be isolated and stored in a freezer for about one week using the developed method. Optimizing the solvent for environmentally friendly large-scale syntheses revealed that methyl ethyl ketone (MEK) was as effective. In addition, in situ-generated α-bromo-δ-valerolactone was directly converted into a sulfur-substituted functional lactone without difficulty by reacting it with a sulfur nucleophile in one pot without isolation. This new bromination system is expected to facilitate the industrial use of α-bromolactones as important intermediates.

    2,2-Bis(phenylselanyl)-1-(p-tolyl)vinyl 2-oxo-2-(p-tolyl)acetate was synthesized via the reaction of p-tolylacetylene with diphenyl diselenide and benzoyl peroxide in benzene under atmospheric conditions. The molecular structure of the synthesized compound was evaluated using single-crystal X-ray analysis and spectral analyses. The process reported here provides a rare example of the direct and selective transformation of a terminal alkyne to the corresponding geminal diseleno-substituted alkene.

    A catalytic oxidation reaction for Acid Blue 7 dye synthesis was evaluated in water. Without lead oxide or manganese oxide derivatives as oxidants, polyoxometalate catalysts were investigated to reduce the usage of harmful heavy metal. A catalyst was prepared by mixing silicotungstic acid with copper oxide, and aqueous hydrogen peroxide (30%) was used as an oxidizing agent. This reaction proceeded to produce Acid Blue 7 from the corresponding leuco acid after 45 min at 95 °C and was viable for a 10 g-scale synthesis.

    The transition-metal-catalyzed diarylation of isocyanides with triarylbismuthines was investigated in detail, and rhodium catalysts such as [RhCl(nbd)]2 were found to selectively afford N-alkyl diaryl ketimines. On the other hand, palladium-catalyzed diarylation proceeded with the incorporation of two molecules of isocyanide, preferentially yielding N,N’-dialkyl or N,N’-diaryl α-diimines. In addition, a cascade synthesis of 2,3-diarylquinoxalines starting from the palladium-catalyzed diarylation of isocyanides with triarylbismuthines was successfully achieved.

    The homolytic cleavage of the P<sup>V</sup>(O)–P<sup>III</sup> bond in tetraphenyldiphosphine monoxide simultaneously provides both pentavalent and trivalent phosphorus-centered radicals with different reactivities. The method using V-40 as an initiator is successfully investigated for the regio- and stereoselective phosphinylphosphination of terminal alkynes giving the corresponding <italic>trans</italic>-isomers of 1-diphenylphosphinyl-2-diphenylthiophosphinyl-1-alkenes in good yields. The protocol can be applied to a wide variety of terminal alkynes including both alkyl- and arylalkynes.

    This study aims to develop green and sustainable advanced synthetic methods based on redox cyclization. Elemental sulfur is shown to promote the efficient synthesis of 2-substituted benzothiazoles from 2,2′-dinitrodiphenyl disulfides and benzylamines via redox cyclization in the absence of transition-metal catalysts and solvents. The 2-substituted benzothiazoles are obtained in good to excellent yields. This synthetic methodology is highly atom-economical and does not require any external oxidizing and/or reducing agents.

    In 2015, the Japanese health insurance approved the use of a second-generation photodynamic therapy (PDT) using talaporfin sodium (TS); however, its cancer cell selectivity and antitumor effects of TS PDT are not comprehensive. The Warburg effect describes the elevated rate of glycolysis in cancer cells, despite the presence of sufficient oxygen. Because cancer cells absorb considerable amounts of glucose, they are visible using positron emission tomography (PET). We developed a third-generation PDT based on the Warburg effect by synthesizing novel photosensitizers (PSs) in the form of sugar-conjugated chlorins. Glucose-conjugated (tetrafluorophenyl) chlorin (G-chlorin) PDT revealed significantly stronger antitumor effects than TS PDT and induced immunogenic cell death (ICD). ICD induced by PDT enhances cancer immunity, and a combination therapy of PDT and immune checkpoint blockers is expected to synergize antitumor effects. Mannose-conjugated (tetrafluorophenyl) chlorin (M-chlorin) PDT, which targets cancer cells and tumor-associated macrophages (TAMs), also shows strong antitumor effects. Finally, we synthesized a glucose-conjugated chlorin e6 (SC-N003HP) that showed 10,000–50,000 times stronger antitumor effects than TS (IC50) in vitro, and it was rapidly metabolized and excreted. In this review, we discuss the potential and the future of next-generation cancer cell-selective PDT and describe three types of sugar-conjugated PSs expected to be clinically developed in the future.

    Chlorin e6 is a heterocycle exhibiting spectral absorption in the 600-700 nm wavelength range used for photodynamic therapy (PDT). Herein, a sugar-conjugated chlorin e6 derivative was synthesized on a large scale. An alkyl spacer was fabricated by controlling the alkoxylation conditions between the thio-sugar and chlorin e6 and thio-glucose-conjugated chlorin e6 was successfully synthesized.

    Greener and more sustainable chemical processes are required to address increasing environmental pollution and depletion of natural resources. This paper aims to develop greener and more sustainable modern synthetic chemical processes using redox-neutral cyclization. Redox-neutral cyclization has been shown to promote the efficient synthesis of 2-substituted benzothiazoles from easily available nitrobenzenes, methyl-heteroaryl compounds, and elemental sulfur in the absence of transition-metal catalysts. The 2-substituted benzothiazoles were obtained in reasonable yields through the sulfuration of electron-deficient C-H bonds with elemental sulfur. This synthetic methodology also affords a high atom economy without the use of any external oxidizing and/or reducing reagents.

    A photosensitizer is a molecular drug for photodynamic diagnosis and photodynamic therapy (PDT) against cancer. Many studies have developed photosensitizers, but improvements in their cost, efficacy, and side effects are needed for better PDT of patients. In the present study, we developed a novel photosensitizer β-mannose-conjugated chlorin e6 (β-M-Ce6) and investigated its PDT effects in human glioblastoma U251 cells. U251 cells were incubated with β-M-Ce6, followed by laser irradiation. Cell viability was determined using the Cell Counting Kit-8 assay. The PDT effects of β-M-Ce6 were compared with those of talaporfin sodium (TS) and our previously reported photosensitizer β-glucose-conjugated chlorin e6 (β-G-Ce6). Cellular uptake of each photosensitizer and subcellular distribution were analyzed by fluorescence microscopy. β-M-Ce6 showed 1000× more potent PDT effects than those of TS, and these were similar to those of β-G-Ce6. β-M-Ce6 accumulation in U251 cells was much faster than TS accumulation and distributed to several organelles such as the Golgi apparatus, mitochondria, and lysosomes. This rapid cellular uptake was inhibited by low temperature, which suggested that β-M-Ce6 uptake uses biological machinery. β-M-Ce6 showed potent PDT anti-cancer effects compared with clinically approved TS, which is a possible candidate as a next generation photosensitizer in cancer therapy.

    Iodine is an element that exhibits characteristic features of heavy halogen, and several compounds containing iodine constitute important synthetic intermediates due to synthetically easy manipulation. To utilize iodine units for organic synthesis, a highly regio- and stereoselective introduction of iodine to versatile building blocks is significant, and a lot of researches for the selective introduction of iodine to alkynes or alkenes have been investigated. In this review article, we describe regio- and stereoselective hydroiodination to multiple bonds of building blocks, and its synthetic applications as key intermediates to construct several important compounds in organic chemistry.

    BACKGROUND/AIM: We previously synthesized a glucose-conjugated chlorin compound e6 (G-chlorin e6), and reported that it has very strong antitumor effects. The aim of the present study was to synthesize acetylated glucose-conjugated chlorin (AcN003HP) and evaluate its antitumor effect and excretion. MATERIALS AND METHODS: To evaluate the antitumor effect of AcN003HP, its IC50 was calculated as well as its accumulation in cancer cells was examined by flow cytometry. Confocal microscopy was used to observe the intracellular localization of AcN003HP. The excretion and antitumor effects of AcN003HP were also evaluated in vivo. RESULTS: AcN003HP showed stronger antitumor effects and accumulation into cancer cells compared to talaporfin sodium, a conventional photosensitizer. AcN003HP was localized in the endoplasmic reticulum. In a xenograft tumor mouse model, AcN003HP showed longer excretion time from the body than G-chlorin e6, and photodynamic therapy using AcN003HP showed very strong antitumor effects. CONCLUSION: The safety, improved controllability, and robust antitumor effects suggest AcN003HP as a good next-generation photosensitizer.

By using the Warburg effect-a phenomenon where tumors consume higher glucose levels than normal cells-on cancer cells to enhance the effect of photodynamic therapy (PDT), we developed a new photosensitizer, glucose-conjugated chlorin e6 (G-Ce6). We analyzed the efficacy of PDT with G-Ce6 against canine mammary carcinoma (CMC) in vitro and in vivo. The pharmacokinetics of G-Ce6 at 2, 5, and 20 mg/kg was examined in normal dogs, whereas its intracellular localization, concentration, and photodynamic effects were investigated in vitro using CMC cells (SNP cells). G-Ce6 (10 mg/kg) was administered in vivo at 5 min or 3 h before laser irradiation to SNP tumor-bearing murine models. The in vitro study revealed that G-Ce6 was mainly localized to the lysosomes. Cell viability decreased in a G-Ce6 concentration- and light intensity-dependent manner in the PDT group. Cell death induced by PDT with G-Ce6 was not inhibited by an apoptosis inhibitor. In the in vivo study, 5-min-interval PDT exhibited greater effects than 3-h-interval PDT. The mean maximum blood concentration and half-life of G-Ce6 (2 mg/kg) were 15.19 ± 4.44 µg/mL and 3.02 ± 0.58 h, respectively. Thus, 5-min-interval PDT with G-Ce6 was considered effective against CMC.

Tetraethylene glycol (TEG)-conjugated (C60-Ih)[5,6]fullerene (TEG-C60) was synthesized as a soluble molecular building block generating C60-based conjugates with advanced properties. The improved solubility allowed the glycosylation reaction with TEG-C60 using a Lewis acid in chloroform at low temperature. The resultant glycoconjugated TEG-C60 was characterized in terms of water solubility, ability to generate reactive oxygen species (ROS), and photodynamic activity.

    がん治療における長年の夢は,腫瘍を非侵襲的に治療することにある.近年光線力学療法(Photodynamic therapy:PDT)が,低侵襲性の治療法として注目されている.我々はPDT用の光増感剤の生体適合性と腫瘍選択性の向上を目指して,光増感剤(クロリン,C60等)への糖質(グルコース,マンノース,マルトトリオース等)の導入を立案した.なかでもグルコース連結クロリンe6(G-Ce6)は,in vitroおよびin vivoで優れたPDT効果を示した.さらに,がん発症犬に対して極めて良好なPDT効果を示した.(著者抄録)
