    胚幹(ES)細胞におけるgene targetingによりSemaphorin 3F(Sema 3F)ノックアウト(KO)マウスを作成し、Sema 3Fの生理学的役割について調べた。その結果、Sema 3F発現の欠損はneuropilin-2(Npn-2)や他の推定Npn-2リガンド、主にSema 3B、Sema 3CあるいはSema 3GのmRNA発現に有意に影響しなかった。Sema 3F KOマウスにおいて体性感覚皮質の樽状構造及び小脳神経解剖は有意に変化しなかった。背側嗅球の固有シグナルの光学的イメージングは、野生型マウスとSema 3F KOマウスの間でodor mapにおいて有意差はなかった。以上より、Sema 3Fはその受容体発現と脳構造の生後発達においてかなり制限された役割を持つことが示唆された。

    短時間の感覚入力を長時間維持される脳活動へと変換するメカニズムは、ワーキングメモリなどさまざまな脳機能に必要とされる。このような感覚入力の維持に、中枢性ニューロンの持続的活動が重要な役割を果たすと考えられているが、そのメカニズムは不明である。本研究では、シンプルな脳構造をもつ嗅覚系をモデル系として、嗅覚一次中枢の投射ニューロンで観察される持続的活動を生成するメカニズムを検証した。本研究により、投射ニューロンの樹状突起間の相互作用を介して匂い信号がアクティブに維持される神経回路メカニズムが新規に見出された。

    嗅覚一次中枢の嗅球において、外界の匂い情報は空間的な「におい地図」として表現されている。脳はにおい地図の情報をどのように解釈し、匂いで惹起される行動反応へと結びつけるのだろうか?嗅球背側部から高次中枢へと向かう嗅覚神経回路が除去された変異マウスでは、天敵などの匂いに対する生得的な恐怖行動反応が消失していた。一方、条件付けによって後天的に獲得される恐怖反応は保存されていた。以上から、本能的な恐怖反応と後天的に学習される恐怖反応にそれぞれ結びつく、異なる嗅覚神経回路が存在することが明らかになった。
