    L-ホモセリン(L-Hse)とNAD(P)Hからなる非リガンド性Thermus thermophilusのホモセリン・デヒドロゲナーゼ(TtHSD)、ならびにLys99AlaとLys195Alaが変異したTtHSDの結晶構造を、それぞれ1.83、2.00、1.87および1.93Åで解析した。L-HseとNADPHの結合はTtHSDの構造をopenからclosedに変化させ、Glu180を含む可動ループが近づいて両者を固定した。Lys99AlaとLys195AlaはいずれもL-Hseの水酸基と水素結合を形成した。TtHSD三重複合体は、closed型の方がopen型より良好にMichaelis複合体を模倣した。Lys99Ala TtHSDの結晶構造はLys99関連水素結合に関与する水分子を含むが、Lys195とL-Hseの位置は維持されていた。以上より、Lys99がHSDの酸-塩基触媒の残基である可能性が考えられた。

    <p>ADP-Ribose pyrophosphatase (ADPRase) reaction was observed in crystalline-state at atomic resolutions around 1 Å. ADPRase achieves the general acid-base catalysis in the presence of divalent metal cations. Crystals of ADPRase alone, ADPRase-ADPR binary (ES-state) complex were prepared, and reaction intermediates were captured by the cryo-trapping after the soaking of MnCl<sub>2</sub> into ES-state crystals. Just after two metal binding sites were successively occupied along with the soaking time, ADPR was hydrolyzed and reaction products were released, indicating the completion of the crystalline-state reaction. The key structures of ADPRase-ADPR-two Mn<sup>2+</sup> ions complex revealed the various enzymatic phenomena including electrostatic, proximity and orientation effects and preferential binding for the transition states. The details of the metal assisting mechanism and the general base catalysis have been discussed.</p>

    Homoserine dehydrogenase catalyzes the conversion of L-aspartate-4-semialdehyde to L-homoserine. Homoserine dehydrogenase is required for the biosynthesis of the three essential amino acids, i.e. lysine, methionine, and isoleucine, from aspartic acid. This enzyme attracts attention also as a promising antifungal drug target. We have determined the crystal structures of homoserine dehydrogenase from Thermus thermophilus HB8 in both substrate-free form and homoserine-binding form by X-ray diffraction. Crystallization conditions were surveyed at 293 K by using a hanging-drop vapor-diffusion method. The substrate-free form was solved to 1.4 Å resolution and the homoserine-binding form was solved to 2.0 Å resolution. The homoserine dehydrogenase was a dimer with each subunit composed of three distinct domains, nucleotide-binding, dimerization, and substrate-binding domain. In the homoserine-binding forms, the amino group of homoserine made a hydrogen bond to the side chain carboxylate of Glu180. This hydrogen bond induced the conformation change of Thr165-Pro183 loop, which made the loop close to the active site. The substrate homoserine was rigidly recognized by several amino acid residues in the enzyme active site, indicating that Lys99 or Lys195 would be an essential catalytic residue to facilitate hydride transfer.

    A new family of NAD(P)H-dependent oxidoreductases is now recognized as a protein family distinct from conventional Rossmann-fold proteins. Numerous putative proteins belonging to the family have been annotated as malate dehydrogenase (MDH) or lactate dehydrogenase (LDH) according to the previous classification as type-2 malate/L-lactate dehydrogenases. However, recent biochemical and genetic studies have revealed that the protein family consists of a wide variety of enzymes with unique catalytic activities other than MDH or LDH activity. Based on their sequence homologies and plausible functions, the family proteins can be grouped into eight clades. This classification would be useful for reliable functional annotation of the new family of NAD(P)H-dependent oxidoreductases.
