    <p>Methods to control the particle sizes of active pharmaceutical ingredients were investigated using sodium ecabet (Na-ECA) hydrate, which tends to generate large plate-like crystals, as a model compound. In batch-cooling crystallization in H<sub>2</sub>O, the particle size decreased as the cooling rate was increased. Although the particle size reached 97 µm at a cooling rate of 40°C/h, this may be too large for formulation without further physical processing. A series of semi-batch crystallization experiments was conducted by adding aqueous Na-ECA solution to aqueous NaCl. In these experiments, the particle sizes decreased to less than 7 µm as the NaCl concentration was increased to >1.2% w/w. The particles yielded by semi-batch crystallization showed improved particle size distributions compared to those obtained by the batch-cooling crystallization method. The solubility of Na-ECA hydrate decreased to less than 10% of that in pure water as the NaCl concentration was increased to 3.0% w/w. This solubility change enabled the production of a highly supersaturated environment, allowing facile generation of microcrystals with a narrow particle size distribution.</p>

    <p>Poly-gamma-glutamic acid (γ-PGA) is a water-soluble, nontoxic biodegradable polymer. It is extensively utilized in medicines, foodstuffs, and cosmetics, as well as in water treatment. Highly pure γ-PGA is required for various purposes. In many cases, γ-PGA is produced by microbes of <i>Bacillus</i> sp. However, an increase in the viscosity of the culture broth is one of the major problems of γ-PGA production, as higher viscosity makes the separation and purification steps difficult. Herein, we propose a novel method to obtain pure γ-PGA using gemini quaternary ammonium salts (GQASs). The quaternary ammonium cation of GQASs binds to the carboxylic acid group of γ-PGA via an ionic bond, following which water-insoluble and antimicrobial complexes are formed. These complexes are obtained in the aqueous solution, which were resolved in ethanol solution. With an increase in the added amount of GQAS, the negative charge of the complex decreased in the aqueous solution. Subsequently, the GQAS within the complexes was dissociated by the addition of NaCl, affording pure γ-PGA in a high yield. Moreover, the complexes were shown to have antibacterial activity and adhered to glass, indicating that the complex itself has a utility value.</p>

    <p>The crystallization mechanism of amino acid derivative <i>t</i>-butoxycarbonyl-L-asparagine (Boc-Asn) was investigated. NMR spectroscopy was used to observe the behavior of Boc-Asn molecules in under- and supersaturated solution. Nuclear Overhauser Effect (NOE) NMR measurements were performed to provide information concerning the intra- or intermolecular access of a particular hydrogen atom to other hydrogen atoms in solution. We compared the interactions identified by NOE measurements for under- and supersaturated solutions of Boc-Asn with those in the crystal, as determined by X-ray analysis. According to the X-ray analysis, intramolecular NOE should not be observed, but intermolecular NOE might be observed between the methyl protons of the <i>t</i>-butoxycarbonyl group and the protons belonging to other functional groups. The time course of the NOE intensity was followed for three solutions with different saturation ratios <i>S</i> (=concentration of solute/solubility). For undersaturated and nearly saturated conditions (<i>S</i>=0.8 and 1.12 at 0°C), the NOE intensity did not change for at least for 250 h. However, for the supersaturated solution (<i>S</i>=1.62 at 10°C), the NOE intensity increased suddenly and rapidly after about 120 h, and no crystals were observed during the NOE measurements. As the observed NOEs agreed with those expected from the arrangement of molecules in the crystal, it was suggested that Boc-Asn molecules aggregated after about 120 h with the same structure as that in the crystal. In addition to the behavior of the Boc-Asn solution before nucleation, we also investigated the effect of aggregation on the crystallization process. Solutions that were preincubated at 10°C for various periods were further cooled and crystallized at −30°C. It was found that when the solution was preincubated under the supersaturated condition for a long time before crystallization, nucleation was enhanced, and consequently, the crystal size distribution became homogeneous.</p>

    <p>To promote the use of sewage sludge on farmland, a flocculant compatible with agricultural application is required. In general, an inorganic flocculant such as aluminium sulfate is used at large sewage disposal plants. However, there is concern that aluminium accumulates when it is returned to farmland, and therefore a biodegradable rather than an inorganic flocculant is required. To obtain biodegradable flocculants produced by microorganisms, the latter were isolated from soils and sewage sludges, and the flocculation activity of the culture broths was evaluated. Here, real sewage sludge was used for the evaluation because practical use was regarded as an important consideration. As a result, two strains, V13 and RK14, were selected and identified as <i>Bacillus megaterium</i> and <i>Bacillus licheniformis</i>, respectively. It was also found that flocculation was caused by poly-γ-glutamic acid (γ-PGA). γ-PGA from RK14 cells had considerably higher molecular weight than commercial γ-PGA, and the enantiomeric composition differed from that of γ-PGA from other <i>B. licheniformis</i>. To obtain mutants with high γ-PGA productivity, RK14 cells were treated with ethyl methanesulfonate, and a mutant (RK14-46) was selected. The culture medium suitable for γ-PGA production by RK14-46 cells was totally different from that by RK-14 cells. The γ-PGA production reached 21.1 g/L when RK14-46 cells were cultured in the optimum semi-synthetic medium at 30°C for 2 d. Furthermore, the combination of γ-PGA obtained from RK14-46 cells and chitosan, a cationic flocculant, was very effective for flocculation of real sewage sludge.</p>

    A mL-scale continuous crystallizer has been newly developed to produce small crystals with a narrow size distribution. The crystallizer is used for the poor solvent crystallization of glycine and <small>L</small>-alanine. The crystallizer is composed of a stainless-steel mixing vessel with an inner volume of 0.9 mL and a high-speed agitator that can agitate the crystallization solution up to 24,000 rpm. Glycine or <small>L</small>-alanine aqueous solution and poor solvent (methanol) are supplied into the vessel at a constant flow rate and intensely mixed by the agitator. The average residence time was set at 0.33, 3.3 and 33 s by changing the flow rate of an amino acid solution and poor solvent. The crystals obtained by the crystallizer are small and uniform in size, comparing with those obtained by a conventional beaker-scale semi-batch and continuous MSMPR type crystallizers. For example, glycine crystals having the 1/38 of the size of crystals obtained by a semi batch crystallizer were obtained. The short residence time that can be attained by the present mL-scale crystallizer is advantageous for the production of small crystals with a narrow size distribution. The size distribution can be further controlled by changing the mixing ratio of poor solvent.

    Nowadays, bio-fuel cells which utilize the enzymes of microbes are widely noticed. In this study, the authors made a bio-fuel cell which utilized dry yeast along with sucrose as fuel, and conducted experiments under various conditions in order to improve its power generation performance. The kinds and concentrations of the electron mediator and oxidizer solutions and the number of current collectors in the anode and cathode were optimized. As a result, the single bio-fuel cell with six sheets of current collectors (each area was 20 cm<SUP>2</SUP>) in each of the anode and cathode was able to generate the maximum power of about 9 mW. Moreover, the authors measured the performance of the fuel cell using wild yeast and analyzed the variations in the concentrations of reaction substances, and further conducted experiments using reagents affecting the metabolism of yeast in order to examine the electricity generation mechanism in the bio-fuel cell.

    The detailed properties of the enzyme from <i>Pseudomonas aeruginosa</i>, which catalyzes the <i>N</i>-acyl linkage between myristic acid and the <i>N</i>-terminal glycine residue of the octapeptide GNAAAARR-NH<sub>2</sub> (PKA) in aqueous solution without ATP and CoA, were studied. The substrate specificity for the acyl peptide in the synthetic reaction was examined, and it was found that at least eight amino acid residues are required for the reaction and that the <i>N</i>-terminal glycine residue is not absolutely essential for the reaction because the activity was detected using the octapeptide that has an <i>N</i>-terminal alanine. The activity was also strongly affected by the amino acid sequence because the activity was very weak in the reaction using GARASVLS-NH<sub>2</sub> (HIV-1p17<sup>gag</sup>). The substrate specificity for fatty acids was also examined. In the reactions using lauric acid and decanoic acid, only slight activities were detected; however, those activities were very small compared with the activity in the reaction using myristic acid. In addition, the degradation of myristoyl PKA by the enzyme was detected, although there are only a few reports on demyristoylation. The optimum pH and temperature of the degradation reaction were consistent with those of the synthetic reaction. The degradation reaction was inhibited by divalent cations.<br>

    The deletion of MCD4 leads to an increase in β-1,6-glucan level and a decrease in glycosylphos-phatidylinositol-anchored protein and mannan levels in the cell wall of Saccharomyces cerevisiae, suggesting that mcd4 deletion mutant(mcd4Δ) displays β-glucans on the cell surface without a mannan cover. An observation of the cell surface of mcd4Δ cells and an examination of the effect of contact between mcd4Δ cells and mouse macrophages indicated that macrophages were activated by contact with mcd4Δ cells displaying β-glucans on the cell surface. We further examined the effect of intraperitoneal ethanol-fixed mcd4Δ cells on the survival period of mice infected with Candida albicans. mcd4Δ cells prolonged the survival period, implying that mcd4Δ cells may enhance the immune function of mice via macrophage activation. Moreover, we examined the structures of β-glucans(i.e., alkali- and acetic acid-insoluble β-glucans) extracted from mcd4Δ with ^<13>C-NMR and the effect of extracted β-glucans on TNF-α secretion from macrophages. The structures of the β-glucans from mcd4Δ differed from those of wild type(WT); however, there was no difference in tumor necrosis factor-α(TNF-α) secretion level between β-glucans from mcd4Δ and those from WT. The yield of purified β-glucans obtained from dry cells of mcd4Δ was higher than that obtained from dry cells of WT. mcd4Δ may be a superior strain for the preparation of β-glucans.

    A water-soluble color indicator was developed for the effective screening of hydrogen-producing microorganisms. This indicator consists of a coloring agent and a water-soluble derivative of Wilkinson's catalyst. Wilkinson's catalyst, Tris(triphenylphosphine) rhodium chloride, had been developed as a catalyst for the hydrogenation of olefins. We used a sulfonate of the catalyst for the hydrogenation of coloring agent in an aqueous medium. Several coloring agents, such as methyl orange, methyl red sodium, neutral red and Evan's blue, dissolved in water together with the sulfonated catalyst showed a change in color when hydrogen gas was fed into the solution by sparging at room temperature. We confirmed that methyl orange was decolorized by biologically produced hydrogen, when the photosynthetic bacterial strain Rhodobacter capsulatus ST-410 was grown in a medium containing 0.6mM catalyst and 0.075mM methyl orange in test tubes of 5ml working volume.

    The enzyme that catalyzes <i>N</i>-acyl linkage between myristic acid and the NH<sub>2</sub>-terminal glycine residue of the octapeptide Gly-Asn-Ala-Ala-Ala-Ala-Arg-Arg-NH<sub>2</sub> in aqueous solution without ATP and coenzyme A was found in <i>Pseudomonas aeruginosa</i>. The enzyme was purified from cell-free crude extract using DEAE-Cellulose, Sephadex G-200, CM-Sephadex C-50, and hydroxyapatite column chromatographies, and then purified approximately 1900-fold with about 1.5% recovery of enzyme activity from the crude extract. Finally, the purified enzyme showed a main band on SDS polyacrylamide gel electrophoresis after staining with Coomassie Brilliant Blue. The band corresponded to a molecular mass of approximately 60 kDa. The <i>K</i><sub>m</sub>s of the purified enzyme for the substrate myristic acid and the octapeptide were 0.36 and 2.6 mM, respectively. When myristoyl-CoA instead of myristic acid was used as the substrate for the enzyme reaction, myristoyl octapeptide could be synthesized as observed in the case of myristic acid. The <i>K</i><sub>m</sub> of myristoyl-CoA was 0.17 mM.<br>

    Most proteins involved in the synthesis of the GPI core structure of Saccharomyces cerevisiae are essential for growth. To explore the relationship between the GPI anchor structure and β-1,6-glucan synthesis, we screened deletion mutants in genes involved in GPI synthesis for osmotic remedial growth. Heterozygous diploid strains were dissected on medium with osmotic support and slow growth of the mcd4 deletion mutant was observed. The mcd4 mutant showed abnormal morphology and cell aggregation, and was hypersensitive to SDS, hygromycin B and K1 killer toxin. Incorporation of GPI cell wall proteins was examined using a GPI-Flo1 fusion protein. The result suggested that the mcd4 deletion causes a decrease in GPI cell wall proteins levels. The mutation also caused a decrease in mannan levels and an increase in alkali-insoluble β-1,6-glucan and chitin levels in the cell wall.
