Research subject summary

  • 1.Elucidating the Molecular Basis of Nutrient-Our research investigates how nutrients—including amino acids, carbohydrates, lipids, vitamins, and minerals—regulate cellular differentiation and metabolic homeostasis through translational control and intracellular signaling. In particular, we focus on how branched-chain amino acid (BCAA) deficiency triggers translational stress responses and modulates the Wnt/β-catenin pathway, aiming to elucidate the molecular mechanisms by which cells sense and adapt to nutritional cues.

  • 2.Analyzing the Mechanisms of Metabolic Regulation by Food-Derived Bioactive Compounds
    We are investigating how food-derived bioactive compounds, such as lactoferrin from milk and curcumin from turmeric, modulate lipid metabolism, inflammation, and ferroptosis sensitivity at the molecular level. By elucidating how these compounds affect oxidative stress responses and lipid homeostasis, we aim to establish a scientific basis for non-pharmacological, nutrition-based interventions against obesity and dyslipidemia.

  • 3.Integrating Nutrient Response Networks for Molecular Nutrition-Based Applications
    We integrate molecular biology and nutritional approaches to construct a comprehensive model of the “nutritional response network,” which describes how nutrients and food-derived compounds interact with cellular molecules. By dynamically analyzing the effects of these factors on gene transcription, post-transcriptional modification, translation, and translational control, we aim to elucidate the molecular principles underlying nutrition-dependent regulation of cellular functions, ultimately establishing a foundation for diet-based strategies to promote health and longevity.

    Apolipoprotein E (apoE) polymorphism is associated with onset of Alzheimer's disease (AD). We found seven polymorphisms in apoE receptor 2 (ApoER2), an apoE-binding receptor, in Japanese sporadic AD patients, but no association of ApoER2 polymorphisms with AD. We consider that the functions of ApoER2 in the brain may be compensated for by those of other apoE-binding receptors such as VLDL receptor.

    The mammalian cellular retinoic acid-binding proteins, CRABP-I and CRABP-II, bind retinoic acid which acts as an inducer of differentiation in several biological systems. To investigate a possible role for CRABP-II in bovine adipogenesis, we have cloned bovine CRABP-II cDNA and the coding region for CRABP-I. The predicted amino acid sequences of CRABP-II were highly conserved among several animal species (human, mouse, and rat at 97%, 93%, and 93%, respectively). The expression pattern of bovine CRABP-II was examined in greater details by applying RT-PCR to various bovine tissues. CRABP-II mRNA was expressed in most adipose-containing tissues. Moreover, the expression of CRABP-I and -II mRNA dramatically increased during the differentiation of adipocytes from bovine intramuscular fibroblast-like cells. The effects of retinoic acid on adipocyte differentiation of bovine intramuscular fibroblast-like cells were concentration-dependent. Retinoic acid activated the formation of lipid droplets at a level of 1 nM, whereas inhibition was observed at a level of 1 μΜ. CRABP-I gene was up-regulated and CRABP-II gene down-regulated by retinoic acid during adipocyte differentiation. These results suggest that CRABPs may play an important role in the regulation of intracellular retinoic acid concentrations during adipogenesis.

    By expression cloning using fluorescent-labeled high density lipoprotein (HDL), we isolated two clones that conferred the cell surface binding of HDL. Nucleotide sequence of the two clones revealed that one corresponds to scavenger receptor class B, type 1 (SRBI) and the other encoded a novel protein with 228 amino acids. The primary structure of the newly identified HDL-binding protein resembles GPI-anchored proteins consisting of an N-terminal signal sequence, an acidic region with a cluster of aspartate and glutamate residues, an Ly-6 motif highly conserved among the lymphocyte antigen family, and a C-terminal hydrophobic region. This newly identified HDL-binding protein designated GPI-anchored HDL-binding protein 1 (GPI-HBP1), was susceptible to phosphatidylinositol-specific phospholipase C treatment and binds HDL with high affinity (calculated K-d = 2-3 mug/ml). Similar to SRBI, GPI-HBP1 mediates selective lipid uptake but not the protein component of HDL. Among various ligands for SRBI, HDL was most preferentially bound to GPI-HBP1. In contrast to SRBI, GPI-HBP1 lacked HDL-dependent cholesterol efflux. The GPI-HBP1 transcripts were detected with the highest levels in heart and, to a much lesser extent, in lung and liver. In situ hybridization revealed the accumulation of GPI-HBP1 transcripts in cardiac muscle cells, hepatic Kupffer cells and sinusoidal endothelium, and bronchial epithelium and alveolar macrophages in the lung.

    セレノプロテインPは主に肝臓で産生される分泌タンパク質であり,セレン輸送タンパク質として機能する。セレノプロテインPの産生は耐糖能異常者や2型糖尿病の患者において増加する。セレノプロテインPの過多な産生はインスリン抵抗性を惹起し2型糖尿病発症の一因であることが示され,必須微量元素セレンの医科学的な重要性が再認識されている。本稿では,グルコース代謝におけるセレノプロテインPの機能とその受容体として機能するリポタンパク質受容体ファミリーについて紹介する。(著者抄録)

    齢や栄養バランスの乱れにより、骨密度低下や筋量減少、代謝異常が進行することが知られています。
    近年の研究から、バリン(Val)やイソロイシン(Ile)などの分岐鎖アミノ酸(BCAA)が、単なるエネルギー源ではなく、細胞内で「栄養シグナル」として遺伝子発現や分化を制御していることが明らかになりつつあります。
    本研究では、BCAA欠乏時に活性化する翻訳ストレス応答やWnt/β-カテニン経路、mTOR/GCN2経路の変化を統合的に解析し、栄養素が骨形成や代謝恒常性をどのように制御するかの分子基盤を解明します。
