    均一な遺伝的背景を持つアスパルトアシラーゼ(Aspa)-em34Kyo/過分極活性化環状ヌクレオチド依存性カリウムチャネル1(Hcn1)-em1Kyo二重ノックアウトラットを作成し、本態性振戦(ET)を抑制するグルタミン酸受容体アンタゴニストの作用を解析した。このラットは、前記の二重変異ラットと比較して同等の自発的振戦を発現した。振戦はN-メチル-D-アスパラギン酸(NMDA)受容体アンタゴニストによって抑制された。非NMDAグルタミン酸受容体アンタゴニストNBQXは振戦を弱く阻害し、代謝調節型グルタミン酸受容体アンタゴニストLY341495は作用しなかった。Aspa/Hcn1二重ノックアウトラットの振戦発症にはイオンチャネルグルタミン酸受容体、特にNMDA受容体が関与すると考えられた。

    ビタミンEは主に抗酸化作用および抗炎症作用を介して動脈硬化・血管障害に抑制的に働く.そこで日本人閉経後女性における動脈硬化性疾患リスクに対する血清ビタミンE濃度の関係を横断的に検討した.閉経後女性107名(70.0±7.7歳)を対象に,血清ビタミンα-およびγ-トコフェロール(αTおよびγT)の測定を行った.動脈硬化性疾患リスク因子数は,血圧高値,高血糖,脂質異常(基準値はメタボリックシンドロームの判定に基づく)の合計数により評価をした.対象者の血清αT濃度,γT濃度の中央値は,24.32μmol/L,2.79μmol/Lであった.ビタミンE欠乏症として溶血性貧血を対象とした場合のαT欠乏の基準値(<12μmol/L)を下回る者はいなかった.血清αTおよびγT濃度の粗値では動脈硬化性疾患リスク数の違いによる有意差は認められなかったが,血清総コレステロール濃度およびトリグリセリド値で補正を行ったαT濃度において,リスク数の増加に伴い,有意な低下が認められた.なお,年齢,BMI,喫煙・飲酒の有無で調整した場合にもこの関係は維持された.以上より,血清ビタミンE濃度は動脈硬化性疾患リスク数に関係し,動脈硬化の抑制を介して生活習慣病の予防に寄与することが考えられた.(著者抄録)
