    <p>　We measured the serum activities of the bone-specific isoenzyme of alkaline phosphatase (ALP3) and tartrate-resistant acid phosphatase 5b (TRAP5b), as osteoblastic bone formation and osteoclastic bone resorption markers, respectively, in two zoo-managed Asian elephant calves during the 4-month period around weaning (2 months before and after weaning). The serum ALP3 and TRAP5b levels decreased after weaning; the levels were significantly higher in the pre-than post-weaning period. These results suggest that osteoblastic bone formation and osteoclastic bone resorption slowed after weaning. Breast milk feeding of Asian elephants may play a significant role in osteoblastic and osteoclastic bone metabolism during the growth period.</p>

    <p>Serum levels of creatine kinase (CK) and lactate dehydrogenase (LDH) isoenzymes were evaluated in nine zoo-managed Asian elephants (<i>Elephas maximus</i>) using a commercial agarose gel electrophoresis (AGE) kit. CK was separated into two major fractions, CK-BB and CK-MM, along with a small fraction of macroenzyme-CK type 2 (mCK2); CK-MM was the largest fraction. LDH was separated into five fractions (LDH1–5); LDH3 was the largest fraction. Age was negatively and positively correlated with the percentages of CK-BB and CK-MM, respectively, and negatively correlated with CK-BB and mCK2 activities. These results indicate that an AGE kit can be used to evaluate CK and LDH isoenzymes. Routine isoenzyme testing may enable early detection of disease and physiological changes.</p>

  • Prepartum measurement of serum biomarkers reflecting osteoclastic and osteoblastic bone metabolism for predicting the risk of milk fever in dairy cows.

    We investigated whether prepartum levels of serum bone biomarkers are related to the degree of parturient hypocalcaemia and risk of milk fever (MF) in dairy cows with advancing parity. A total of 58 late-pregnant cattle were assigned to four groups: nulliparous, primiparous, multiparous in the 2nd lactation and multiparous in the 3rd-5th lactation. The multiparous cows were further assigned to MF and non-MF groups according to the onset of MF. Serum samples were obtained from the cows during the 3 weeks prepartum to 5 d postpartum period for the measurement of serum calcium (Ca) and three bone biomarkers: tartrate-resistant acid phosphatase isoform 5b (TRAP5b), osteoprotegerin (OPG) and bone isoenzyme of alkaline phosphatase (ALP3). The ratios of OPG to TRAP5b (O/T ratio) and ALP3 to TRAP5b (A/T ratio) were calculated. The data from all cattle showed that the severity of hypocalcaemia at parturition increased with advancing parity/age. The MF cows had elevated serum TRAP5b activity and a decreased O/T ratio after parturition, suggesting an increased number of osteoclasts due to osteoclastogenesis, in response to severe hypocalcaemia. The MF cows showed lower serum ALP3 activity during the 3 weeks prepartum than the non-MF cows, therefore, prepartum osteoblast function was likely weak in the MF cows. During the 2-3 weeks prepartum, serum ALP3 activity and the A/T ratio had moderate associations with the serum Ca concentration at day 0 (day of calving) in the multiparous cows, and receiver operating characteristic curve analysis revealed that ALP3 activity had excellent ability to predict MF. In conclusion, prepartum serum ALP3 activity is a promising biomarker to predict MF in multiparous cows.

  • The effects of maternal supplementation of rumen-protected lysine during the close-up dry period on newborn metabolism and growth in Holstein calves

    This study aimed to evaluate the effects of rumen-protected lysine (RPL) supplementation during the close-up period on blood metabolites and calf growth. Forty multiparous Holstein dams were selected based on parity, body condition score, and expected calving date, and randomly assigned to a group: with RPL (n = 22) or without (control [CON], n = 18). RPL dams were supplied daily with 80 g of RPL from Day 21 before the expected calving date to parturition. Blood samples were obtained from the dams before the start of supplementation, 1 week before calving, and immediately after calving, and from calves immediately after birth and weekly until 8 weeks of age. Body weight measurements were performed immediately after birth in all calves and at weekly intervals until 8 weeks of age in female calves. No significant difference was observed in serum metabolite levels and plasma amino acid concentrations between the RPL and CON dams before supplementation, whereas plasma lysine concentrations tended to be higher in RPL dams immediately after calving (p = 0.07). Serum total protein levels (p < 0.05) were higher, whereas plasma total amino acid, total essential amino acid, total non-essential amino acid, and other amino acid concentrations were lower in the calves of RPL dams than those of CON dams (p < 0.05). There were no significant differences in calf birth weight between the two groups, although female calves of RPL dams (n = 7) had higher serum total protein (p < 0.05) and tended to have greater body weight (p = 0.09) from 1 to 8 weeks of age than those of CON dams (n = 11). Overall, RPL supplementation during the close-up period may increase placenta-mediated amino acid transfer to the foetus and enhance protein synthesis in the calf, leading to improved weight gain during the suckling period.

    <p>The approved Japanese measurement method of circulating alkaline phosphatase (ALP) has changed from that of the Japan Society of Clinical Chemistry (JSCC) to that of the International Federation of Clinical Chemistry and Laboratory Medicine (IFCC). We measured the serum levels of total ALP (t-ALP) and those of the isoenzymes ALP2 and ALP3 in 50 Asian elephant (<i>Elephas maximus</i>) specimens using both methods. The activities determined by the IFCC method were roughly one-third lower than those determined by the JSCC method. We present conversion formulae. Our results enable comparisons of historical and current data on serum ALP activities in endangered, zoo-managed Asian elephants.</p>

  • 貫通固定ピンキャスト(TPC)で治療したホルスタイン育成牛の橈尺骨骨折の1例

    ホルスタイン種の雌育成牛(238日齢、体重247kg)における左橈尺骨骨幹の非開放性斜骨折と診断した症例について、貫通固定ピンとフルリムキャストを用いた貫通固定ピンキャスト(TPC)法による治療を行った。術中は塩酸キシラジンの静脈内投与(0.2mg/kg)で仰臥位保定後、塩酸メデトミジンの定量持続点滴(20μg/kg/hr)にて鎮静を維持し、2%リドカイン10mlによる腕神経叢ブロックで疼痛管理した。患肢の牽引と骨把持鉗子により骨折端を合わせて固定し、整形外科用パワードリルを用いて直径4.0mmの貫通固定ピンを、骨折端を挟む遠位および近位側橈骨骨幹にピンを各2本、近位および遠位骨の骨折端および骨折面を貫通するようにピン3本をそれぞれ貫通させた。さらにその上からフルリムキャストを巻き、キャスティングテープ表面から露出したピンは約4cmで切断し、即時重合レジンで各ピンの切断部を被覆・結合した。手術時間は80分であった。症例は覚醒後すぐに起立歩行可能であり同日中に退院、単房で個別飼育とし、術後1週間の抗生物質製剤投与を行った。術後48日目にX線所見にて明瞭な仮骨形成を認めたため、手根関節以下の遠位側のキャスティングテープ部分を切断・除去した。術後81日目にキシラジン鎮静下でTPC全てを除去し治癒とした。治療期間中および治療後の起立歩行は問題なく、その後の発育も順調であった。牛の橈骨骨折では内固定で高い治癒率が報告されているが治療にかかるコスト、手術時間および難易度、術後合併症のリスクにおいてTPCは内固定よりも生産動物である本症例にメリットを与えた。牛の骨折治療では体重が予後に影響を与え、さらにTPCでは使用する貫通固定ピンの太さ・本数・配置ならびにフレームであるキャストの長さが、骨折端の安定性や固定の強度に影響を与える。体重が重い個体の橈骨骨折をTPCにて治療した報告は限られており、本症例は今後の治療の際の良いモデルとなった。(著者抄録)

    乾乳牛へのペレット状グリセリン給与が分娩前後の栄養代謝状態と分娩後の疾病発生や生産性へ及ぼす影響を調査した。分娩予定3週前から分娩までグリセリン80g含有ペレット(Gly、20頭)またはプラセボペレット(Cnt、21頭)を毎日給与した。Cntのルーメンフィルスコアは分娩前に低下したが(P<0.05)、Glyで変化せず、分娩前の血中β-ヒドロキシ酪酸(BHBA)濃度はGlyで低かった(P<0.05)。分娩前後のボディコンディションスコア(BCS)はGlyで高かった(前;P<0.05、後;P=0.06)。ケトーシス発症頭数に差はないが治療牛の血中BHBA濃度はGlyで低い傾向にあった(P=0.07)。以上より、分娩予定3週前からのグリセリンペレット給与は給与中の採食量を一定に保ち脂質代謝を改善させ、分娩前後のBCSの適切な維持やケトーシスの症状を緩和させる可能性が示された。(著者抄録)

    Dairy cattle management lacks consideration of fetal breed, the effect of which on fetal growth and nutrition are unclear. We investigated blood parameters in 12 late-pregnant Holstein heifers with similar (Holstein, n = 5) or different (Japanese Black [n = 4] or F1 cross [n = 3]; Holstein × Japanese Black) fetus breeds and in their umbilical cords and calves. Samples were obtained from dams 1 week before calving (-1 week) and immediately after calving, from the umbilical vein at calving, and from calves immediately after birth. Dams with beef fetuses had higher serum glucose levels (-1 week; p < .05) than those with Holstein fetuses. Plasma total amino acid, total essential amino acid, total nonessential amino acid, and other amino acid concentrations were lower in the umbilical veins of dams with calves of the beef breeds than in those of the Holstein breeds (p < .05). Furthermore, serum glucose and plasma amino acid levels were lower in the beef calves than in the Holstein calves (p < .05). Overall, nutrient supply from dams to beef fetuses was lower than that to Holstein fetuses. Our findings may facilitate feeding management of dairy cattle pregnant with beef breeds for appropriate fetal growth and nutrition.

    This study investigated consecutive fluctuations in serum activities of bone-specific alkaline phosphatase (ALP) isoenzyme 3 (ALP3) in 11 clinically healthy Holstein heifers during the first 18 months of life. ALP3 activities at the first sampling time point after weaning (3 months) were significantly lower than those at multiple time points during the pre-weaning period. Those activities increased from a minimum at 3 months to a peak at 6 months during the post-weaning period. In the anthropometric data, daily body weight and wither height gains appeared to be below the public data at 4 months and 4-5 months, respectively. The data suggested that serum ALP3 activity can be used to monitor skeletal growth of heifers at weaning.

    This study investigated changes in serum levels of hepatic, bone, and intestinal alkaline phosphatase (ALP) isoenzymes (ALP2, ALP3, and ALP5, respectively) in Holstein cows around parturition. Tartrate-resistant acid phosphatase 5b (TRAP5b) activity and calcium (Ca) concen-trations were also measured. We analyzed blood samples from 11 late-pregnant heifers (primipa-rous group) and 13 multiparous (2-4 lactations; multiparous group) cows at 3 weeks (18-24 days prepartum; -3 weeks), 2 weeks (17-11 days prepartum; -2 weeks), and 1 week (10-4 days prepar-tum; -1 weeks) before parturition; the day of calving (within 12 h post-calving; day 0); and 5 days postpartum (5 days). ALP3 activity was significantly higher in the primiparous group than in the multiparous group, whereas the activities decreased significantly in both groups after 5 days. ALP2 and ALP5 activities did not change, whereas ALP2 activity was significantly higher in the primiparous group than in the multiparous group. TRAP5b activity was significantly higher in the primiparous group than in the multiparous group and showed a transient significant increase at day 0. Ca concentration significantly decreased at day 0 in both groups; the Ca level at day 0 was significantly higher in the primiparous group than in the multiparous group. These data show that ALP3 activity in serum may indicate a change in osteoblastic bone forma-tion after calving, but further study is needed to determine the clinical application for measuring ALP isoenzymes in bovine medicine.

    Osteoprotegerin (OPG) inhibits osteoclast (OC) differentiation. TRAP5b (tartrate-resistant acid phosphatase 5b) secreted by OCs reflects the numbers of mature OCs. This study assessed these OC-related markers around parturition in cows of different parities and in cows with milk fever (MF). The blood OPG and TRAP5b concentrations, as well as the ratio of OPG to TRAP5b (O/T), were measured beginning 3 weeks before (-21 d) and over a few days after calving in 49 Holstein Friesian cows at first (n = 8), second (n = 17), third (n = 12), and fourth or greater (n = 12) parities. The ratio of O/T at -21 d to O/T at calving (PreCOT) was also calculated. In the third and greater parities, seven cows developed MF (non-MF, n = 17). Regardless of the development of MF, the serum OPG started to decline in the last week of gestation only in the cows entering the second lactation, while the blood TRAP5b increased at calving in all cows. O/T decreased toward parturition only in multiparous cows. The decrease in O/T at caving was less pronounced in MF cows. PreCOT was negatively correlated with lactation number only in multiparous cows (n = 41, ρ = -0.50, P < .01). This study implied that OC differentiation toward calving was tapered in cows with advanced parities, and these indexes predict the risk of MF.

    国内の肉牛生産現場において、去勢は最も頻繁に行われる外科処置の1つである。去勢は生産性向上の観点から畜産現場で広く実施され、なかでも観血法による精巣摘出が主流とされている。今回、観血去勢後の手術部位感染(surgical site infection:SSI)により陰嚢膿瘍を形成した黒毛和種去勢牛3頭に対して、一般身体検査および超音波診断装置を用いて陰嚢や精索の形状、膿様物の有無を精査した後、外科的治療を行った。局所的な陰嚢膿瘍と診断した2例において、膿瘍は非感染部との境界が明瞭であったことから陰嚢の全切除を行った。1例において精索は膿瘍として大きく腫大したまま腹腔内深部へと連絡していたことから、陰嚢切除ならびに精索瘻による腹腔内膿瘍の排膿処置を行った。術後には継続的な精索瘻の内腔の洗浄を行い、精索の大きさは退縮、排膿の量も減少、手術後42日目には精索瘻の炎症消退と創傷収縮を認めた。いずれの症例も治癒転帰となったことから、陰嚢切除ならびに精索瘻による腹腔内膿瘍の排膿は有効な治療法であったと考えられた。今回の3例は、獣医師ならびに畜産関係者に対して、簡易外科手技である去勢術の衛生的な手技実施ならびにSSI予防対策に努めることの必要を再認識させる重要な症例であった。(著者抄録)

    The anthropometric and blood data of an unsuccessfully hand-reared Asian elephant (Elephas maximus) calf were retrospectively compared with the data for calves raised by their real mothers or allomothers, to identify potential reasons for poor outcomes in the hand-reared case. The hand-reared calf grew normally in terms of body weight and withers height. However, blood biochemical data suggested reduced bone metabolism, low immune status, and malnutrition during its life. Blood bone markers were measured to determine whether a skeletal disorder was present in the Asian elephant calf, which was not clear from the anthropometric data. Monitoring these parameters in hand-reared Asian elephant calves, with the aim of keeping them within the normal range, may increase the success rate of hand-rearing of Asian elephant calves.

    The purpose of this study was to elucidate the colostral and foal serum immunoglobulin G (IgG) concentration values in heavy draft horses in Japan and to examine the effects of peripartum mare condition on colostral immunity. Colostrum was obtained 1 hr after foaling (pre-suckling; n=178). Blood was collected from the jugular vein of the foals (n=147) at 24 to 48 hr after birth. The foaling statuses of 73 mares were recorded. The average colostral IgG concentration was 10,540 ± 3,190 mg/dl (median=10,928; range 1,434-17,514 mg/dl). The average serum IgG concentration obtained from neonatal foals 24 to 48 hr after birth was 1,750 ± 919 mg/dl (median=1,890; range 0-3,510 mg/dl). Although colostral IgG did not differ between the normal foaling mare (n=59) and dystocial mare (n=14), foal serum IgG was lower in foals born in dystocia than in foals in normal foaling (P<0.05). This study demonstrates reference values for colostral and foal serum IgG specific to heavy draft horses in Japan and suggests that dystocia may interfere with the acquisition of colostral immunity in neonatal foals.

    A recent study found that an agarose gel electrophoresis (AGE) method yielded two distinct major bands corresponding to the hepatic and bone ALP isoenzymes (ALP2 and ALP3, respec-tively) in bovine serum treated with protease and neuraminidase (PN-treatment), although there were concerns that the intestinal ALP isoenzyme (ALP5) often overlapped with ALP3 in human serum treated with neuraminidase. Because ALP5 was separated from ALP3 in bovine serum treated with protease alone (P-treatment), we used a modified method employing both P- and PN-treated bovine sera to measure the activities of the three ALP isoenzymes in 53 lacta-ting Holstein cows: 24 primiparous and 29 multiparous. Upon electrophoresis, 51 of 53 samples (96.2%) subjected to P-treatment yielded a distinct fraction corresponding to ALP5, as did the control serum. All PN-treated sera yielded a definite ALP2 fraction. The ALP3 fraction was calculated as the remainder after excluding ALP2 and ALP5. The activities of total ALP (t-ALP) and ALP3 in primiparous cows were higher than those in multiparous cows (p ⟨ 0.001) at early-to-peak [10-110 days in milk (DIM)] and mid (111-220 DIM) lactation. In the multi-parous cows, the ALP3 activity at late lactation (221-477 DIM) was significantly higher than that at early-to-peak lactation. Thus, the modified AGE method described here is able to discrimi-nate three fractions of ALP isoenzymes in the sera of lactating cows. The AGE pattern of circu-lating ALP isoenzymes will contribute to the understanding of the physiological bone metabolism status in lactating cows.

  • Intestinal ganglioneuroma in a neonatal Japanese black calf - Short communication.

    A 1-day-old male calf presented with clinical signs of severe progressive abdominal distension. Abdominal radiographic and ultrasonic images revealed several loop-like structures in the small intestine, which were indicative of gas retention. Experimental laparotomy was performed. However, the calf died during surgery. At necropsy, a round, well-circumscribed mass (3 × 3 × 2.5 cm) was found in the jejunal wall, and the jejunal lumen was narrowed. The mass was firm and had white to grey appearance on the cut surface. Histologically, the submucosa and the muscle layer were diffusely thickened due to abundance of neural tissues comprising several fascicles of nerve fibres and large aggregates of ganglion cells. Some ganglion cells contained basophilic Nissl substances in their cytoplasm. Immunohistochemically, these cells were positive for S-100 and synaptophysin. Ultrastructural examination revealed that the neoplastic ganglion cells contained dense core vesicles in the cytoplasm. Based on these findings, the neoplastic lesion was diagnosed as ganglioneuroma in the jejunum.

    Liver performs several important functions; however, predicting its functions is difficult. Methods of analyzing gene expression profiles, for example, microarray, provide functional information of tissues. Liver and peripheral blood leukocytes (PBLs) were collected from Holstein cows subjected to two different physiological conditions (non-pregnant and pregnant), and PBLs were fractionated by gradient cell separation. RNA from PBLs and liver were applied to oligo-DNA microarray and reverse transcription-quantitative polymerase chain reaction (RT-qPCR). It revealed a group of stable bovine liver genes under constant physiological conditions. When they applied to physiological conditions including non-pregnant and pregnant, the profiles of some genes in liver were consistent with those in PBLs. Microarray data subjected to a principal component analysis (PCA) showed that the hepatic gene expression profiles were more consistent with those of granulocytes than mononuclear cells. The relationship of gene profiles in liver with granulocytes was confirmed by RT-qPCR and hierarchical cluster analysis. Gene profiles of granulocytes were more reliable than those of mononuclear cells, which reflected liver functions. These results suggest that the genes expressed in PBLs, particularly granulocytes, may be convenient bioindicators for the diagnosis of clinical disorder and/or detecting aberration of liver functions in cows subjected to different physiological conditions.

    We measured the bone-specific alkaline phosphatase (ALP) isoenzyme activity in 67 plasma samples from 14 newborn Holstein calves using both a conventional method (featuring heat inactivation) and a commercial agarose gel electrophoresis (AGE) kit; the relevant isoenzymes were termed bone-specific ALP (BAP) and ALP isoenzyme 3 (ALP3). We explored whether the AGE kit afforded reliable data when used to analyze samples from Holstein calves. The blood was collected from the jugular vein of each calf immediately prior to the first colostrum feeding (pre-feeding), 20 and 40 h after pre-feeding, and on days 4 and 7; whereas three samples (from three calves) were not obtained. The total plasma ALP activity varied widely, exceeding the ranges of reference values. On electrophoresis, 52 of 67 plasma samples (77.6 %) clearly contained both ALP isoenzyme 2 and ALP3, as did control human serum. The total ALP activity of the 52 samples ranged from 166-1989 U/L (median: 1013 U/L), whereas the values for the other 15 samples (22.4%) exhibiting abnormal isoenzyme fractionation ranged from 1014-5118 U/L (median: 1780 U/L). In the 52 plasma samples exhibiting clearly separated isoenzymes, ALP3 and BAP activities were strongly positively correlated as revealed by Deming regression (y = 0.93x + 22.6, p⟨0.0001) and Bland-Altman analysis (ALP3/BAP activities limit of agreement: -5.1%). Thus, the AGE kit yields useful information on newborn calves, and can replace the conventional method when the total plasma ALP activity is less than approximately 1000 U/L.

    To clarify the possibility of Mycoplasma mastitis in milking cows from Mycoplasma pneumonia outbreaks in calves, Mycoplasma bovis was isolated over time from nasal swabs of calves and bulk tank milk at two dairy farms where no cases of Mycoplasma mastitis occurred. Isolates were genotypically analyzed using pulsed-field gel electrophoresis. At both farms, Mycoplasma bovis was first detected in bulk tank milk after pneumonia outbreaks in calves, and the genotypes of the isolates from nasal swabs and milk were consistent. These findings indicate that outbreaks of Mycoplasma pneumonia in calves is one of the possible etiologies of Mycoplasma mastitis in dairy herds.

    The aim of this study was to examine the treatment outcomes of tibial fracture using different configurations of transfixation pinning and casting (TPC) in eight cattle. After pins were inserted to stabilize the fracture site, cast material was placed from the level of the stifle to the hoof (full TPC) in six cases and from the stifle to the proximal metatarsus (partial TPC) in two cases. Five cattle (three full TPC and two partial TPC cases) underwent TPC removal 36-86 days after surgery and resumed productivity. The other three cattle which underwent full TPC suffered from irreversible orthopedic complications within 5 weeks of surgery, while a prolonged healing process was observed in the partial TPC cases. Although TPC is an effective method for treating bovine tibial fracture, full TPC may result in a guarded prognosis in some cattle. However, partial TPC can delay healing. Surgeons should choose the optimal TPC configuration while considering the various factors that affect the healing process.

    The bone-specific alkaline phosphatase (ALP) isoenzyme activity was measured in 51 serum samples from four captive Asian elephants (Elephas maximus) using a conventional method with wheat germ lectin precipitation and a commercial agarose gel electrophoresis (AGE) kit; the isoenzymes were designated as bone-specific ALP (BAP) and ALP isoenzyme 3 (ALP3), respectively. This study examined the suitability of the AGE kit for analyzing blood biochemistry in Asian elephants. The serum ALP3 and BAP activities were strongly positively correlated and met the evaluation criteria for agreement using Bland-Altman analysis. The results indicate that the AGE kit can be used to examine the blood biochemistry in Asian elephants instead of the conventional method.

    The aim of this study was to elucidate the bone metabolic status after taking colostrum in newborn calves. Fourteen neonatal calves were randomly allocated to two groups fed either unheated or heated (60°C, 30 min) colostrum three times on the first day (2 l every 10 hr; 6 l in total). Heat treatment on colostrum was to reduce the bone metabolic markers assumed as heat-sensitive. The concentrations of four bone metabolic markers (the enzymes from bone cells or the bone collagen fragments) and a bone protective protein, osteoprotegerin (OPG), were measured in the blood of calves during a week after the birth and in the colostrum. The colostral concentrations of four bone metabolic markers were reduced by heating. Then those circulatory markers peaked after colostrum intake in the calves fed unheated colostrum; whereas those fed heated colostrum did not show such changes. However, the plasma tartrate resistant acid phosphatase 5b (TRAP5b) activity was transiently increased after taking colostrum in both groups. Meanwhile, heating did not decrease colostral OPG and there was no significant rise in the serum OPG concentrations after the first colostrum intake in both groups. The study revealed that the blood concentrations of studied bone metabolic markers depended on those colostral values except for TRAP5b. Based on the plasma TRAP5b changes, accelerated formation of premature osteoclast cells may be induced by colostrum intake. Meanwhile, colostral OPG absorption is less likely to impact on its circulating levels.

    Instructive information on the treatment for chronic deep infections of digital joints in a 9-month-old Holstein heifer is described in this report. Preoperative ultrasonographic and radiographic examinations revealed soft tissue swelling and subchondral bone lysis at the distal part of the proximal phalanx in the lateral digit. Arthrectomy was performed under xylazine sedation to remove infectious articular cartilage tissues. Immature callus formation was observed via radiography at the surgical site by the 28th postoperative day. On the 48th postoperative day, callus fell into disrepair on the radiographs along with aggravation of the locomotion score. After the application of a half-limb cast, the immature callus formed again by the 62nd postoperative day (11 months), and bony callus formation was observed by the 74th postoperative day. Thereafter, the heifer could walk well with marked improvement in the locomotion score. The withers height of the heifer at 13 months (136 cm) was within the range of that in control heifers of the same age on this farm (133 ± 3 cm); however, the body weight (BW) of this heifer (322 kg) was lower than the BW of controls (384 ± 26 kg). The BW gain from 11 to 13 months of age seemed to be higher in the present heifer (+76 kg) than in controls (+55±20 kg), suggesting that BW of the present heifer was returning to the original BW. Based on these observations, we suggested that arthrectomy was an effective treatment option for the present case of digital joint arthritis.

    ホルスタイン種の雌育成牛(238日齢、体重247kg)における左橈尺骨骨幹の非開放性斜骨折と診断した症例について、貫通固定ピンとフルリムキャストを用いた貫通固定ピンキャスト(TPC)法による治療を行った。術中は塩酸キシラジンの静脈内投与(0.2mg/kg)で仰臥位保定後、塩酸メデトミジンの定量持続点滴(20μg/kg/hr)にて鎮静を維持し、2%リドカイン10mlによる腕神経叢ブロックで疼痛管理した。患肢の牽引と骨把持鉗子により骨折端を合わせて固定し、整形外科用パワードリルを用いて直径4.0mmの貫通固定ピンを、骨折端を挟む遠位および近位側橈骨骨幹にピンを各2本、近位および遠位骨の骨折端および骨折面を貫通するようにピン3本をそれぞれ貫通させた。さらにその上からフルリムキャストを巻き、キャスティングテープ表面から露出したピンは約4cmで切断し、即時重合レジンで各ピンの切断部を被覆・結合した。手術時間は80分であった。症例は覚醒後すぐに起立歩行可能であり同日中に退院、単房で個別飼育とし、術後1週間の抗生物質製剤投与を行った。術後48日目にX線所見にて明瞭な仮骨形成を認めたため、手根関節以下の遠位側のキャスティングテープ部分を切断・除去した。術後81日目にキシラジン鎮静下でTPC全てを除去し治癒とした。治療期間中および治療後の起立歩行は問題なく、その後の発育も順調であった。牛の橈骨骨折では内固定で高い治癒率が報告されているが治療にかかるコスト、手術時間および難易度、術後合併症のリスクにおいてTPCは内固定よりも生産動物である本症例にメリットを与えた。牛の骨折治療では体重が予後に影響を与え、さらにTPCでは使用する貫通固定ピンの太さ・本数・配置ならびにフレームであるキャストの長さが、骨折端の安定性や固定の強度に影響を与える。体重が重い個体の橈骨骨折をTPCにて治療した報告は限られており、本症例は今後の治療の際の良いモデルとなった。(著者抄録)

    乾乳牛へのペレット状グリセリン給与が分娩前後の栄養代謝状態と分娩後の疾病発生や生産性へ及ぼす影響を調査した。分娩予定3週前から分娩までグリセリン80g含有ペレット(Gly、20頭)またはプラセボペレット(Cnt、21頭)を毎日給与した。Cntのルーメンフィルスコアは分娩前に低下したが(P<0.05)、Glyで変化せず、分娩前の血中β-ヒドロキシ酪酸(BHBA)濃度はGlyで低かった(P<0.05)。分娩前後のボディコンディションスコア(BCS)はGlyで高かった(前;P<0.05、後;P=0.06)。ケトーシス発症頭数に差はないが治療牛の血中BHBA濃度はGlyで低い傾向にあった(P=0.07)。以上より、分娩予定3週前からのグリセリンペレット給与は給与中の採食量を一定に保ち脂質代謝を改善させ、分娩前後のBCSの適切な維持やケトーシスの症状を緩和させる可能性が示された。(著者抄録)

    国内の肉牛生産現場において、去勢は最も頻繁に行われる外科処置の1つである。去勢は生産性向上の観点から畜産現場で広く実施され、なかでも観血法による精巣摘出が主流とされている。今回、観血去勢後の手術部位感染(surgical site infection:SSI)により陰嚢膿瘍を形成した黒毛和種去勢牛3頭に対して、一般身体検査および超音波診断装置を用いて陰嚢や精索の形状、膿様物の有無を精査した後、外科的治療を行った。局所的な陰嚢膿瘍と診断した2例において、膿瘍は非感染部との境界が明瞭であったことから陰嚢の全切除を行った。1例において精索は膿瘍として大きく腫大したまま腹腔内深部へと連絡していたことから、陰嚢切除ならびに精索瘻による腹腔内膿瘍の排膿処置を行った。術後には継続的な精索瘻の内腔の洗浄を行い、精索の大きさは退縮、排膿の量も減少、手術後42日目には精索瘻の炎症消退と創傷収縮を認めた。いずれの症例も治癒転帰となったことから、陰嚢切除ならびに精索瘻による腹腔内膿瘍の排膿は有効な治療法であったと考えられた。今回の3例は、獣医師ならびに畜産関係者に対して、簡易外科手技である去勢術の衛生的な手技実施ならびにSSI予防対策に努めることの必要を再認識させる重要な症例であった。(著者抄録)
